| Tripterygium wilfordii Hook. f.(TWHF) is a traditional Chinese medicinal herb, and distributes widely in southern China,such as Fujian,Zhejiang,Anhui,et al. It has been widely used in the clinical treatment of rheumatoid arthritis, other inflammatory and autoimmune diseases and male fertility control. Pharmacology studies have demonstrated that TWHF can treat inflammation, tumors, HIV/AID and Alzheimer's disease. However, TWHF has low therapeutic index, and the incidence of side effects is much higher than that of other plants,so we must be careful when using it.The compounds of TWHF are highly complex,some micro-ingredients have fort-bioactivity. So,for the using safely and effectely in the clinical treatment,we need an effected method to control the quanlity of the TWHF.The Characteristic Chromatogram of Tripterygium wilfordii Hook. f. bioactive component was establishen by HPLC method.We usually use the root without the velamen and the root for the clinical treatment traditionally,but limited by the resouse of the TWHF and researched deeper,the leaves is also used in the treatment.In this research,we mainly studied on the TWHF collected in the GAP basic land from Taining,Fujian, the Characteristic Chromatogram of Tripterygium wilfordii Hook. f. bioactive component was established by HPLC method for the first time which was divided to four parts(the leaves,the velamen,the root core and the stem). The results showed that the methods were stable,reliable,repeatable and specific for analysis content of bioactive components from the four parts of TWHF,and the Characteristic Chromatogram by these methods were systemly,wholely and characteristic.The results showed that the contents from the four parts had various differences among them,the velamen have a largest number of the components,and the contents differ greatly among them;the root core have less components than the velamen,which coincides with the view of "used to use the root without the velament clinically",but the contents of them vary with the season,and it was the most at autumn, simultaneously,the contents were higher collected in the GAP basic land from Taining than the wilding,so we should grow the TWHF canonical normatively;When we use the TWHF,we often discard the stem of it, which is a very large resource-wasting,the result of this research showed that the contents of the components in the stem were stable and a great variety,so we can develop and use the stem of the TWHF;The content of triptolide in the leaves of the TWHF were most at autumn,and the shatter have the similar components,which suggested that we could develop and use the shatter of the TWHF.In sum,we cannot control the quality of the TWHF and the preparations only by control some one component.It provided the basic theory for the deep research of the TWHF.The methods can be used to evaluate the quality of the different parts of TWHF effectively,and it also provided the technical support and the fundamental researches data for the further rational utilization and the Drug-using safety clinically, effectively and controlly.The methods can be applied to the quality control of Tripterygium wilfordii Hook. f..In addition,as a further research,the relationship between the bioactive components group and their effects,was necessary and important.The method of content determination was established by chromatographic experiments.The result showed that the method was easy to operate and accurate.The content of triptolide from TWHF was determined by HPLC method.The results showed that the contents of triptolide from TWHF in different producing time and parts revealed a sharp difference:there was a lower content in the leaves of TWHF collected in spring than the velamen,and higher than the velamen collected in winter. The results showed that we must collect the TWHF regelized and avoid to the waste of the TWHF, and the results also indicated we can use the leaves of TWHF in the clinical treatment, which provided the basic theory for the better use of the TWHF. |
