Experimental Study Of Therapeutic And Preventive Effects Of Bushen Huoxue Fang On OA Cartilage Degeneration Of The Knee

Objective:To explore the effects of Bushen Huoxue Fang on degenerated cartilage and cell apoptosis in OA animal model in rats.Methods:40 SD rats were divided randomly into control group, OA group, Bushen Huoxue Fang treated group, HA treated group and NS treated group (55101010 Rats for each group). OA animal model was made in rats by the way of cutting off the anterior cruciate ligament, removing the medial meniscus in last four groups.4 weeks later, the fore 2 groups rats were killed, observing the pathology change of cartilage and synovium to check whether the animal model is successfully set up.0.1 ml traditional Chinese drug extract was injected into the knee joints in C group,0.4 mg/kg HA in D group and 0.1 ml saline in the E group once a week, continue for 5 weeks. After 2 months of the first injuction,the last 3 groups rats were killed.The proximal tibia articular cartilage were examined for the degeneration and cell apoptosis of cartilage and synovium.The index of cell apoptosis also is measured.Results:(1)Macroscopic observation and Light microscope results showed that, compared with group A, group B showed a significantly degenerated cartilage.lt means that the OA animal model is successfully produced. (2) Light microscope results showed that it was some improved in cartilage shap. Mankin's score and Apoptosis cell index have significant difference between C,D group and E group(P<0.01)Conclusions:It suggests that the Bushen Huoxue Fang has some protective effect on KOA cartilage degeneration and cellapoptosis.
