| Objective:In recent years, the incidence of intervertebral disc degeneration disease is gradually increasing, which brings a lot of inconvenience to the people’s life. "BuShen HuoXue ShuJin" decoction clinical treatment has great curative effect on intervertebral disc degeneration. To study the effect of "BuShen HuoXue ShuJin" decoction on the expression of heat-shock protein in the treatment of degenerative intervertebral disc disease, searching the therapeutic effectiveness mechanism of "BuShen HuoXue ShuJin" decoction.Methods:45 male SD rats selected, aged 12-13 weeks, weight of about 320-400g, randomly divided into 3 groups,15 rats in each group, those are the control group (group A), blank control group (group B), Chinese medicine group (group C). Using a needle which use a 18G needle limited and 5mm long prick the tail intervertebral disc 6/7,7/8 of the rat. After one month, four rat are selected from group B and group C to conduct MRI detection, and observe the change of Intervertebral disc. Other rats carry out the corresponding treatment. Group A and group B are feed with normal saline one month, Group C is feed with "BuShen HuoXue ShuJin" decoction one month. After the end of the irrigation, taken out intervertebral disc under the anesthesia. Using Immunohistochemical method detect and compare HSP27, HSP90β expression.Results:Group B, C and group A conduct the observation of MRI imaging. It comes to the conclusion that the Intervertebral discs of two animal model groups expression obvious degeneration. Obvious degeneration of rat tail intervertebral disc can be seen in visible model group. The expression of HSP27 in group A, group B and group C has statistical significance, but the HSP90 β has no statistical significance.Conclusions:Pricking tail intervertebral disc successfully establish of tail disc degeneration mode of the rats. "BuShen HuoXue ShuJin" decotion has effect on HSP27 but not on HSP90β when it treat intervertebral disc degeneration, which the mechanism related to the two proteins must be do further research. |
