Dysfunctional uterine bleeding (DUB) is defined as abnormal uterine bleeding without a demonstrable organic cause. DUB, especially that associated with heavy bleeding, is frequently associated with symptoms such as fatigue, discomfort and depression. Through this project, one or more effects and mechanisms of GXY on Dysfunctional Uterine Bleeding are expected to be developed, main contents of this paper consist of three parts as follow:1. Experimental Study on The Stanching Effect and Mechanism of Dysfunctional Uterine Bleeding by GXYThe incomplete-abortion rat model was induced by misoprostol and mifepristone. Uterine bleeding was collected and determinated the OD value of the wavelength of 546 nm, trial rats in diestrus after the last administration in rat of each group, The thymus, spleen, adrenal gland, uterus and ovary were taken out immediately and calculate viscera index. Light microscopy was used to observe the pathological changes of uterine; Serum E2 and P values were determined by RIA(radioimmunoassay method); uterine ERmRNA and PRmRNA expression were detected by the RT-PCR. The results revealed①GXY(11,22,44 g/kg)could markedly reduce uterine bleeding quantity in uterine bleeding model rats(p<0.05,0.01).②GXY could increase the index of uterus, ovary and the light microscopy showed glands and lamina of uterine decreased.③GXY (22g/kg) could significantly increase the level of serum estradiol (E2) in model rats. But it can not regulate the level of progestogen(P) in this model.④GXY (5.5,11,22g/kg) could significantly decrease the level of estrogen receptor (ER) and progesterone receptor(PR) (P <0.05, P<0.01)of endometrium in model rats.The effects of GXY on clotting and bleeding time of mice were observed with slide method and severed tail. The results revealed GXY(11,22,44 g/kg) could markedly shorten the bleeding time and clotting time.Besides, signal recorder was used to measure the effects of GXY on contractile amplitude and frequency of the isolated normal, oxytocin stimulated uterus from rats. The contractive intensity was increased in normal uterus and decreased in oxytocin stimulated uterus in vitroGXY can stanch, mitigate the viscera index,uterine morphological changes and can regulate the deranged sexual hormone level and the expression of uterine ERmRNA and PRmRNA in incomplete-abortion induced by medicine in early pregnancy rats. GXY can also regulate contractive function of uterine in normal uterus and decreased in oxytocin stimulated uterus in vitro.2. Influence of GXY on Proliferation and Secretion of Granulosa Cell The 21d rat should be hypodermiced PMSG 66U/n 48h before death, we investigated the growth characteristic of GC isolated from the rat ovary. GC were cultured at 37℃,5%CO2, either alone or in the presence of GXY on the co-culture system of the serum for 48h. The viability of GC was measured by MTT method (490nm) and the estradiol and progesterone levels was measured by radioimmunoassay (RIA) method. GC injury model were induced by cadmium chloride (CdCl2), the viability of GC was measured by MTT method and estradiol and progesterone changes were by RIA. The results revealed GXY could increase the number and viability of GC (p<0.01), promote the secretion of estradiol and progesterone of GC. In addition, GXY had a protective effect of injured GC and to promote the secretion of estradiol(E2) and progesterone (P).3. Experimental Study on Analgesia and Anti-inflammation Effect of GXYThe effects of GXY on analgesia was observed on the models of the oxytocin and acetic acid-induced writhing. The results revealed GXY culd obviously decrease the writhing times of the oxytocin and acetic acid-induced writhing and inhibited the auricular edema, exudation of abdominal blood capillaries.The anti-inflammatory actions of GXY were observed on the models of xylene-induced ears swelling and acetic acid-induced capillary permeability increase. The results revealed GXY could obviously inhibited the auricular edema, exudation of abdominal blood capillariesGXY showed the effect in the oxytocin and the acetic acid-induced writhing in mice, and showed anti-inflammatory effect in the xylene induced mice ears swelling and in acetic acid induced increased vas cular permeability tests in mice.
