| Purpose: The different molecular weight fragments of Zymosan is prepared from the natural product of Zymosan through separation and purification, and these samples are carried on CAG activity research, and the constructed relation is researched.Method: First Zymosan is isolated and purified by hydroxylation, ultrafiltration and dialysis, and then the total carbohydrates, uronic acids and protein is assayed by the phenol-H2SO4, phloroglucinol method and BCA kit for protein quantification method; And the purity and the distribution of molecular weight is determined by HPLC; The component of sugars is determined by PMP and GC; Also the mode of connection between sugars is determined by GC-MS .And last the activeness the samples is confirmed and the construction effect relations is deduced according to CAG model of mouse by pharmacodynamics.Result: Three molecular weight fragments of Zymosan 2K, 5K, 8K has been got, respectively the total carbohydrates is 63.17%,75.5%,84.45%, and the content of protein is 9.84%, 5.61%, 1.14%, however the properties of uronic acids is very low. Three samples all include great (1-3)Glu and few (1-3)Man and few (1-6)Glu. And about branching point, 2K is at the 4-position?of(1-6)-Glu, 5K is at the 4-position?of(1-6)-Glu and 6-position?of(1-3)-Glu, 8K is at the 4-position and 6-position of(1-3)-Glu and 4-position of (1-6)-Glu. Three samples are benefited for the treatment of CAG, and the curative effect of 8K molecular weight fragment of Zymosan has the obviously improvement, however 5K and 2k almost have the same curative effect.Conclusion: 2K,5K,8K molecular weight fragments have been got from Zymosan, and they are researched of the different structures and different moleculat weight fragment of Zymosan and CAG model of mouse on the structure-activity relationship, the effect of 8K molecular weight fragment of Zymosan has the significantly improvement。... |
