| Objective:A HPLC method for analyzing the constituents in rat plasma after oral administration of Da Cheng Qi Deccotion (DT), a traditional Chinese medical formula, has been carried out.Methods:The facile and reproducible HPLC fingerprints of the formula in vitro and in vivo were set up. The similarity evaluated by Correlation Coefficient and Principal Component Analysis(PCA) was employed to estimate the consistency and difference of the fingerprints. In addition, the compounds originated from the formula and their metabolites in the plasma were identified by comparing the fingerprints in vitro with that in vivo.Results:19 constituents have been detected in vitro; 8 were identified; 13 constituents have been deteced in vivo and 2 were identified. Compound I and XIII in rat plasma exist originally in Rheum officinale Baill; compound IV and V belong to Citrus aurantium L, and others were the metabolites of Rheum officinale Baill.1.5.10.13 peaks were verified as important contribution to classification by PCA,it it indicated the concentration of these metabolites in vivo change very quickiy,it may be bioactive constituent. Then, quantitative determination of Rhein in it was also developed, It is concluded that a rapid and validated method has been developed based on HPLC-DAD, which shows high sensitivity and resolution that is more suitable for identifying the bioactive constituents in plasma after oral administration of Chinese herbal medicines, and provides helpful chemical information for further pharmacology and active mechanism research on the Chinese medical formulaConclusio:This method of serum pharmacochemistry is applied to research the material foundation of Dachengqi Decoction.
