| Objective Rhubarb,as one of the ancient and important commonly used traditional Chinese medicine in TCM clinic,has long medicinal history and its different processed products were often used in different prescription to play its unique therapeutic effects. The core of TCM processing Principle is the material basis and biological activity changed.This paper,keeping the purgation by bitter-cold of rhubarb as the main line and taking rhubarb, rhubarb wine, cooked rhubarb and rhubarb carbon extracts as the research object,which researched and compared the biological activity and mechanism of different processed products of rhubarb through purgation by bitter-cold, activating blood circulation to dissipate blood stasis and antipyretic effect in order to provide basis for the scientific connotation of potency changed by pcocessing and clinical rational drug use.Method According to purgative ED50 of mice by Bliss, the first purgative time, the 5-hour purgative times and defecation quantity and the small intestinal propulsion in mice in vivo by charcoal powder rates, this experiment observed the different purgative effect among different samples of rhubarb extracts; Investigating the effects of rhubarb extracts on Na+-K+-ATPase in rat colon wall cell membrane to show the purgative effect mechanism.The acute blood stasis model in rats were established by combining the adrenalin and ice-bath.The effects of the processed products of rhubarb on the idexes of hemorheology such as blood viscosity, plasma viscosity packed volume of erythrocyte, plasma fibrinogen levels,the platelet functions such as platelet aggregation and the anticoagulant functions such as PT,TT,PTT were observed.The febrile rats were established by injecting suspension of yeast hypodermically.Then the temperatures at 1,2,4 and 6h after administration were observed, the content of PGE2 and cAMP were measured by Enzyme-Linked Immunosorbent Assay(ELISA) to research the antipyretic effect and mechanism of different processed products of rhubarb.Result The purgative effect:the purgative ED50 of rhubarb was equivalent to the rhubarb wine, the cooked rhubarb and the rhubarb carbon were not purgative, judging from the first purgative time, the 5-hour purgative times and defecation quantity, the purgative effect of rhubarb was greater than rhubarb wine.compared with control group, the rhubarb and rhubarb wine can promote the small intestinal propulsion in mice (P<0.01 or P<0.05), but he cooked rhubarb and rhubarb carbon cann't. Compared with control group, the rhubarb, rhubarb wine, cooked rhubarb and rhubarb carbon extracts can inhibit Na+-K+-ATPase in rat colon wall cell membrane (P<0.01 or P<0.05), but at the same dose the rhubarb has the strongest inhibitory effect, next came the rhubarb wine, cooked rhubarb and rhubarb carbon. To some content, the result of Na+-K+-ATPase was consistent with the purgative effect. The blood circulation promoting and blood stasis removing effect:compared with control group, the idexes of hemorheology of model group were significantly increased(P<0.01 or P<0.05), which showed that the model was successful.compared with model group, the blood viscosity, plasma viscosity, packed volume of erythrocyte and platelet aggregation were significantly decreased than that in rhubarb and rhubarb wine group (P<0.01orP<0.05), the rhubarb wine also can decrease the plasma fibrinogen levels (P<0.01orP<0.05); the cooked rhubarb can decrease the blood viscosity and platelet aggregation (P<0.05); the rhubarb carbon can decrease the plasma fibrinogen levels and the low dose can decrease the blood viscosity (P<0.05). The rhubarb and rhubarb wine can extend PT,TT (P<0.01 orP<0.05), the high dose of the raw rhubarb also extend APTT(P<0.01), but the rhubarb wine not; the cooked rhubarb can extend PT,APTT (P<0.05); the rhubarb carbon can extend PT (P<0.05).The antipyretic effect: compared with model group, the rhubarb and rhubarb wine can decrease the rising body temperature of the febrile rats at every point(P<0.01 or P<0.05); the antipyretic effect of cooked rhubarb and rhubarb carbon was significantly poorer than the rhubarb and rhubarb wine (P<0.01 or P<0.05);compared with model, the rhubarb and rhubarb wine can significantly decrease the hypothalamic PGE2 and cAMP level caused by yeasts(P<0.01 or P<0.05), the cooked rhubarb and rhubarb carbon have a tendency to decrease the hypothalamic PGE2 and cAMP level, but no difference statistically.Conclusion From the diarrhea effect, the raw rhubarb is the strongest, wine rhubarb the second, and the cooked and carbon rhubarb almost have no influence on diarrhea; Rhubarb diarrhea is related to the inhibits of the colon Na+-K+-ATP enzyme. Comprehensively analyzing the idexes of hemorheology and the platelet functions, the raw Rhubarb,the wine rhubarb and the cooked rhubarb has the effect to promot the blood circulation and remove blood stasis, among them, the wine rhubarb has the strongest effect; to some extent, the rhubarb-carbon also show weak effect, and the rhubarb carbon shows a two-way regulating role at different dose. Four kinds of processed products have antipyretic effect in different degree and its mechanism is relevant to the decrease of PGE2 and cAMP levels of the hypothalamus. |
