| Objective:patients with diabetes is growing constantly, Diabetic Angiopathies is a common complication of diabetes, also one of the primary cause lead to the death of diabetic patients. A great quantity of studies show that inflammatory cytokines play a very important role in diabetes and its vascular complications' development。There are also studies found a correlation between hypothyroidism and inflammatory cytokines.The purpose of this study was to investigate the effect of diabetes mellitus with hypothyroidism upon serum level of inflammatory cytokines and related mechanisms.Methods:There are 49 SD Male rats, weight 180g-220g, randomly divided into into A (thyrozolreg gavage group), B (control group) groups, including 26 rats in group of A. Thyrozol 10mg dissolved in 50ml normal saline, A group was given 0.29mg/100 (g.d) orally, B group was given the same amount of normal saline, continuous intragastric administration of 2 weeks, free eating and drinking, taking weight per week, adjusted dose according to the weight. Orbital venous blood tests thyroid function(TT3,TT4 and TSH)before and after intragastric gavage for 2 weeks, comparing with B group, the TT3 and TT4 of group A of two weekend was significantly decreased, and TSH significantly increased as a successful model of hypothyroidism. The 23 rats of group A consistent with hypothyroidism entered the next step. Divided group A rats that is hypothyroidism into hypothyroidism group (S group 10) and hypothyroidism with diabetes group (DS group 13); The 23 rats of group B were divided into control group (N group 10) and diabetes mellitus group (D group 13). The rats of DS group and D group inject the left lower quadrant with 55mg/kg 1%STZ, other groups were given the same amount of citric acid-sodium citrate buffer. Measuring the random blood sugar of caudal vein with One Touch SureStep after 7days, when blood glucose≥13.9mmol/L was identified as a successful diabetic model. Measuring the fasting and random blood sugar of caudal vein with One Touch SureStep Before intragastric gavage,1 weekend and 3 weekend after the diabetic model was successfull made. After Successfully made diabetic model for 3 weeks, rats were fasted for 8 hours, orbital venous blood tests TT3, TT4, TSH, IL-6, hs-CRP, TC, TG, LDL-C, HDL-C and other indicators, All data uses mean±standard deviation (χ+S) to express, and statistical package SPSS 17.0 was used for statistical analysis.Results:The serum level of IL-6 in D group, S group, DS group were significantly higher than in the N group (P<0.01), especially in the DS group, comparing with D group and S group were also significantly higher (P<0.01), however D group and S group had no statistical difference. The serum level of hs-CRP in D group, S group, DS group were significantly higher than in the N group (P<0.01), especially in the DS group, comparing with D group and S group were also significantly higher (P<0.01), however D group and S group had no statistical difference. Doing correlation analysis at the end of the trial (5 weeks) shows that hs-CRP and IL-6, TC, TG, LDL-C, HDL-C, TSH, TT3, TT4 and random blood glucose were correlated (P<0.01), while fasting blood glucose was no correlation (P>0.05). Use hs-CRP as the dependent variable, IL-6, TC, TG, LDL-C, HDL-C, TSH, TT3, TT4 and random blood glucose as independent variables, multi-factor linear stepwise regression analysis hs-CRP and IL-6, TC, TG, LDL-C, HDL-C, TSH, TT3, TT4 and random blood glucose, hs-CRP is mainly determined by IL-6, coefficient of determination R=0.680, standardized regression coefficient is 0.829.Conclusion:The serum level of CRP and IL-6 in Diabetic rats with hypothyroidism was significantly higher than the pure hypothyroidism and pure diabetes; The serum lipid level in diabetic rats with hypothyroidism was significantly higher than pure diabetes and simple hypothyroidism; Multi-factor linear regression analysis confirmed that the level of CRP mainly determined by IL-6. Diabetes, Particularly associated with hyperipidemia should be screened for thyroid function. |
