Objective:To investigate the correlation of HER-2 gene status and BRCAl, Ki-67 gene expression in breast cancer. The development mechanism of breast cancer was analyzed by investigating the relationship between HER-2 gene status and the BRCA1,Kj-67 gene expression of breast cancer and their clinicopathological factors.Methods:HER-2 gene status in 53 casos of breast invasive ductal carcinoma were detected by fluorescencel in situ hybridization. BRCA1,Ki-67 protein expression of them were deteted by immunochemical SP method. The relationships between gene expression and clinicopathological parameters with axillary lymph node metastasis were analyzed in 53 cases of breast invasive ductal carcinoma and 18 cases of breast fibroadenoma tissu.Results:The invasive ductal breast cancer patients of HER-2 gene amplification were BRCA1 gene deletion or decreased expression of protein, Ki-67 protein expression level increased, the difference was statistically significant (P<0.05) Showed HER-2 gene amplification of lymph node-positive was obviously higher than ofI lymph node-negative group, the difference was startistieally significant (P<0.05). BRCA1 in breast invasive ductal carcinoma group was significantly lower than that of breast fibroadenoma group, the difference was statistically significant (P<0.05). Ki-67 in breast invasive ductal carcinoma group was higher than that of breast fibroadenoma group, the difference was statistically significant (P<0.05).Conclusion:The detection of HER-2 gene status and BRCA1, Ki-67 gene expression provide strong scientific evidence for the occurrence and development mechanism of breast cancer BRCA1 in breas invasive ductal carcinoma was significantly lower than that of breast fibroadenoma.Ki-67 in breast invasive ductal carcinoma was higher than that of breast fibroadenoma. In the patients of breast cancer HER-2 gene amplification, BRCAl gene is able to detect absence or decreased expression of protein, While detection of Ki-67 gene levels of protein expression is increased. BRCA1 protein expression and HER-2 gene status was a negative correlation. Ki-67 protein expression and HER-2 gene status was a positive correlation. Results provide scientific basis for early prevention, early diagnosis, reasonable and appropriate treatment by gene targeting and accurate judgement prognosis in breast cancer, So, it has good clinical and pathological value.
