Objective:To detect the single nucleotide polymorphisms of miRNA genes in Hazakh's esophageal carcinoma in XinJiang Province, explorer the relationship between Hazakh's esophageal carcinoma and the single nucleotide polymorphisms of miRNA genes.Methods:Using the matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis to detect the single nucleotide polymorphisms of 266 cases of Hazakh's esophageal squamous cell carcinomas(ESCC),222 cases of cancer-free controls in the same area; All the calculations and statistics were performed with the computer program, SPSS version 17.0 and SHEsis.Results:(1) Kazakh esophageal carcinoma miR34b gene rs 11606481 C/T polymorphisms genotype frequencies were 7.8%,46.1% and 46.1%, there was significant difference compare with the genes of control (14.8%,62.1%,23.1%) (P<0.05); (2) Kazakh esophageal carcinoma miR219-1 gene rs107822 G/A polymorphisms genotype frequencies were 32.5%,49.8% and17.7%, there was significant difference compare with the genes of control (17.5%,48.7%,33.8%), genotype frequencies associated with ESCC pathology differentiation (P<0.05); Kazakh esophageal carcinoma miR219-1 gene rs213210 C/T polymorphisms, there was no significant difference significant compare with the genes of control; (3) Kazakh esophageal carcinoma miR124-1 gene rs531564 C/G polymorphisms genotype frequencies were 68.0%,28.3% and3.7%, there was significant difference compare with the genes of control (58.0%, 39.87%,2.2%); (4) Significant linkage disequilibrium was observed between miR219-1 gene rs107822 and rs213210 polymorphisms, and complete linkage disequilibrium was observed in control group. (5) Four haplotypes were found between miR219-1 gene rs107822 and rs213210 polymorphisms:G,AT,AC,å'ŒGC, and the frequencies of haplotypes were significantly different between ESCC and control group (P<0.05), GT haplotype was associated with a increased risk of ESCC (OR=1.669,95%CI:1.238-2.251), AT haplotype was associated with a reduced risk of ESCC (OR=0.608,95%CI:0.352-0.671); (6)There was no polymorphisms in miR34a in Kazakh race, and the frequencies of miR146a,miR423,miR34b/c,miR100,miR373,miR375 genotypes and alleles were not significantly different between Kazakh ESCC and Kazakh race (P>0.05).Conclusions:(1) miR34b, miR219-1 and miR 124-1 polymorphisms were associated with ESCC risk in Kazakh; (2) Significant linkage disequilibrium was observed between miR219 gene rs 107822 and rs213210 polymorphisms in Kazakh; and the frequencies of haplotypes were associated with ESCC, the GT haplotype may be an harmful factors for ESCC, but AT haplotype may contribute to protection for ESCC.
