| Objective:Detect the methylation conditions of miR-34 gene in Hazakh's esophageal carcinoma in XinJiang Province, to explorer the relationship and significance between Hazakh's esophageal carcinoma and the methylation of miR-34 gene.Methods:Using the matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis to detect the methylation conditions of 145 cases of Hazakh's esophageal squamous cell carcinomas(ESCC),60 cases of non-cancer tissues adjacent to esophageal squamous cell carcinomas(NCAE) and 39 cases of mucous membrane of normal esophagus(NE); Using software of Cluster 3.0,TreeView and GraphPad Prism 5.0 to analysis the distribution of the CpG units among the ESCC,adjacent and NE; Using SPSS 17.0 software package to do all the statistical analysis.Results:(1) our results showed that the methylation of miR-34 gene in ESCC is higher than the NCAE and NE. (2) Comparison of the methylation rate among ESCC,NCAE and NE,there were 5 units(CpG1.2.3, CpG9.10, CpG11.12.13, CPG14, CpG15.16) among the 8 CpG units that could be detected had statistical significance(p<0.05)(3)Comparison of the methylation of different rate among well-differentiated,moderately or poorly differentiated ESCC,the methylation difference (moderately-poorly differentiated> well-differentiated) of miR-34b/c CpG1.2.3 had statistical significance(p<0.05); Comparison of the methylation rate between different TNM staging, the methylation difference (â… +â…¡>â…¢+â…£)of miR-34b/c CpG9.10,CpG11.12.13 had statistical significance(p <0.05);Comparison of the methylation rate between gender,age,position,gross, position,gross,lymph node metastasis and HPV16 infection, there were no CpG unit had statistical significance(p>0.05).Conclusions:(1) the methylation of miR-34 gene in Hazakh's Esophageal squamous cell carcinomas is higher than non-cancer tissues adjacent to tumor and normal control,indicate that mthylation of miR-34 b/c gene may be involved in the pathogenesis of Hazakh's esophageal carcinoma in XinJiang Province.(2) Hypermethylation of miR-34b/c CpG ites CpG1.2.3 maybe correlated with Hazakh's esophageal carcinoma pathology differentiation,higher methylation in poor differentiation. and hypermethylation of CPG9.10,CpG11.12.13 maybe correlated with Hazakh's esophageal carcinoma clinical staging,indicate that involved in the tumor earlay progress. |
