| [Background and purpose]In human tumors, thymoma is relatively rare in the class of tumors.It comes from the thymus, mainly located in the anterior mediastinum.It is more common in adults than in children and the incidence is no difference between men and women Most of thymoma are benign in clinical terms. According to clinical symptoms and signs and imaging examination,the diagnosis of thymoma is not difficult at present. In the treatment of thymoma, surgery is the main way. The prognosis of thymoma is mostly well.The survival rate is relatively high. In recent years,studies have proved that, P53 and Ki-67 in tumor's development, metastasis and prognosis can play an important role. Studies have shown that in the thymoma there is the overexpressions of p53 and Ki-67,suggesting that p53 and Ki-67 in the process of thymoma cell's malignance play an important role. P53 and Ki-67 expression was positively correlated. This study's purpose is to investigate the expression and correlation of P53 and Ki-67 in human thymic tumors,and their possible role in tumor genesis, infiltration and metastasis.[Materials and methods]1 Clinical Materials 50 cases of thymoma specimens for the First Affiliated Hospital of Zhengzhou University from 2005.7 to 2009.6 menstrual pathologically confirmed surgical resection specimens.28 cases were male and 22 cases were female; aged from 2 years to 73 years,with an average age of 38 years.All specimens by histopathological examination were confirmed thymoma organization and were formalin-fixed,paraffin embedding,serial sectioning standby.2 Experimental Methods To use immunohistochemical SP method. P53 mouse anti-human monoclonal antibody, mouse anti-human Ki-67 monoclonal antibody,SP immunohistochemistry kit and dia minobenzidine (DAB) color kit were purchased,FuJian maixin Biotechnology Co. In five cases of normal thymus tissue, Ki-67 is on expression. In 50 cases of the thymic tumors, with the increasing malignant degree of thymoma, the positive rate of Ki-67 were rising (X2=8.452, P=0.015). The positive ratio of Ki-67 were increased fromâ… toâ…¡+â…¢+â…£(x2=4.757;P=0.029).3. P53 positive cases were positively correlated with Ki-67 in thymic tumors.[Conclusion]1. Increased P53 and Ki-67 expression might play an important role in the genesis, invasiveness and metastasis of thymic tumors. 2. Detection of the two protein expressions simultaneously might be more helpful in making an early diagnosis of the tumors judgement of theirs malignant degree, invasiveness and metastasis capacity, as well as the prognosis.3. P53 and Ki-67 showed a positive correlation between the two in the occurrence of thymoma, development and beginning the process of malignant synergy. |
