Astragalus Polysaccharides Treat Type 1 Diabetes Mellitus In Nonobese Diabetic Mice Transplantated By Mesenchymal Stem Cells And Its Possible Mechanisms In Immunology

The Traditional Chinese Medicine plays a important role in many aspects on the transplantation of bone marrow mesenchymal stem cell (MSC), for example,increase the number of MSC, Induced differentiation In vitro, Interfere with the body after the transplantation and so on. The existing experimental reports show that APS can intervent NOD mice to become the model of T1DM, reduce the incidence of T1DM, delay the onset, reduce the degree of insulitis,and protect ofβcell ultrastructure.In the project, it is first to use APS to intervent the MSC transplantation in curing T1DM.This article is divided into two parts.The first part is theoretical study, we firstly elaborate of "the research on diabetes in Traditional Chinese Medicine".Then we get to know the research on T1DM in Modern Medicine.In the other review named The applications of Astragalus Polysaccharides(APS) in the treatment of Diabetes Mellitus; At last,we descript" The theoretical basis on treatment of T1DM by the transplantation of mesenchymal stem cells ".These reviews will be able to make the research better as the basic theory. Also,it provides experimental evidence for APS used for the clinical treatment of T1DM.The second part is experimental study,take spontaneous animal model of T1DM,named NOD/Lt mice,wait until the onset of TIDM,then randomly select 24 mice and divide into 4 groups evenly:Group A,intravenous injection with APS;Group B, Transplantation of Mesenchymal stem cells(msc); Group C, intravenous injection with APS+transplantation of MSC; Group D,blank control.Test tail vein blood glucose and weight weekly;after 3 weeks,kill mice,test serum lymph cell factor, measure regulatory T cells in the spleen content, take Immunohistochemistry of insulin on pancreas. The results show that,APS is a kind of effective immunomodulator,it can lower blood sugar, maintain weight, improve the immune environment, protect the remaining islet cells; it has a positive synergistic effect as the same as transplantation of MSC in Regulation of lymphocyte immune function and Treatment of type 1 diabetes.
