Objective:To investigate the effect of Astragalus Po lysaccharides (APS) combined with Panax Notoginseng saponins (PNS) on kidney of rats with diabetic, and explore the possible mechanism of its function from transforming growth factor beta1, platelet-derived growth factor, type IV collagen and laminin using the method of immunohistoehemistry.Methods.-Diabetic was induced by intraperitoneal injection60mg/kg streptozotoc in on male SD rats. The diabetic rats were randomly divided into model group, APS group (300mg/kg), PNS group (100mg/kg), PNS combined with APS group (APS300mg/kg+PNS100mg/kg), benazepril group (0.9mg/kg). Rats were administered intragastrically one time a day for8weeks. The weight of kidney and body, urinary protein level in24h, blood sugar, blood creatinine and urea nitrogen were determined and the kidney organ coefficient was calculated. The rats were sacrificed and kidney were stained with HE. The protein expression of transforming growth factor beta1(TGF-β1), platelet-derived growth factor(PDGF-BB), type Ⅳ collagen (Col-IV) and laminin (LN) in the renal tissue were determine by immunohistoehemistry.Results:Weight of the rats were significantly reduced (P<0.01); kidney organ coefficient, unrinary protein level in24h, blood glucose, creatinine and urea nitrogen level in rats in the model group were significantly higher than those in normal control group (P<0.01), Histopathological examination showed glomerular basement membrane thickening, tubular appear a large number of inflammatory cell infiltration, interstitial fibrosis. The protein expression of TGF-Pi, PDGF-BB, Col-JV and LN in the renal tissue were enhanced (P<0.01). Compared with the model group, weight of the rats in the APS and PNS group were slightly increased (P<0.01), kidney organ coefficient, unrinary protein level in24h, blood glucose, creatinine and urea nitrogen level in rats were reduced (P<0.05, P<0.01). H istopathological examination showed glomerular basement membrane thickening were lighten, glomerular lesions were reduced. The protein expression of TGF-β1,PDGF-BB, Col-Ⅳ and LN in the renal tissue were waned (P<0.01). Weight of the rats in PNS combined with APS group were obviously increased (P<0.01), kidney organ coefficient, unrinary protein level in24h, blood glucose, creatinine and urea nitrogen level in rats were obviously reduced (P<0.01). Histopathological examination showed glomerular basement membrane thickening were obviously lighten, glomerular lesions were obviously reduced. The protein expression of TGF-β1, PDGF-BB, Col-IV and LN in the renal tissue were waned than the APS group, the PNS group (P<0.01).Conclusion The results showed that APS combined with PNS had protective effect or kidney of rats with diabetic, and better than that of APS and PNS alone. The mechanism may be related to the protein expression of TGF-β1, PDGF-BB, Col-IV and LN.
