Chronic disease is harm to human's health and the quality of life as its high prevalence and mortality rate. With the development of the living standard and the changing of lifestyle of people, chronic diseases, such as heart disease, stroke, cancer, chronic respiratory diseases and diabetes, are more popular and some of them are at low age. So, large medical fees have become a new ponderous burden of individual, family and society. On medical, the root cause of chronic diseases is supposed to be immunity dysfunction of human body. Human are susceptible to various illness due to constant immunodepression caused by bacteria and virus. In other words, the effective method of preventing chronic disease is to enhance human's immunity. And there is an urgent demand for immunoenhancement medicine or health food.Recently, botanical polysaccharides have aroused the most attention, because they are nontoxic and do not cause significant side effects. Polysaccharides are ideal candidates for therapeutics with immunomodulatory. It is reported that there are several different targets of immune modulatory of botanical polysaccharides, such as cellular immunity, humoral immunity and nonspecific immunity. But one kind of plant polysaccharides may only have effects on only one or several targets. In other words, the compound of several polysaccharides may betterly improve the immune function due to their synergistic effect. Based on the reasons above, the immune-enhancing activity of marine compound polysaccharides composed of oligomannuronic acid and polysaccharides from Ganoderma Lucidum was studied.Firstly, the crud polysaccharides from Ganoderma Lucidum were systematically investigated. The enzymatic extraction of crud polysaccharides from Ganoderma lucidum was optimized. And the DPPH free radicals scavenging activity of their ultrafiltration products (GLU-1~GLU-3) was studied. GLU-1 was chosen due to its best antioxidant properties. Different molecular polysaccharides from Ganoderma Lucidum (GLP-1~GLP-8) were gained by low pressure column chromatography. The result of the research in vitro indicated that GLP-7, 5, 6 have more potent antioxidant effect than the others. Then, the structure of GLP-5 was investigated by UV, IR and NMR.In addition, the antioxidant activity of polymannuronic acid (M) and polyglucuronic acid (G) was studied. M was degradated by Fenton Reagent into different low molecular M due to its better antioxidant activity. And M3000 was chosen by antioxidant activity experiment as one of the materials of marine compound polysaccharides.The marine compound polysaccharides was composed of GLU-1 and M3000. And the scanning activities of the three kind of free radicals (DPPH?,?OH, O2-?) of GLU-1, M3000 and marine compound polysaccharides (GLU-1+M3000 1:1) were detected. The result showed that all of them were active for oxidation resistance, and the clearance rate is as follows: GLU-1+M3000 (1:1) > M3000> GLU-1. That is to say, the marine compound polysaccharides inhibited the best antioxidant activity. And this study provides necessary data for its further investigation.At the same time, the immune-enhancing activity of GLU-1, M3000 and marine compound polysaccharides (GLU-1+M3000 1:1) were investigated. The result showed that, comparing with the blank group, compound polysaccharides significantly promoted the thymus/body weight ratio (P<0.01), the level of serum hemolysin (P<0.01), DTH (P<0.01) and enhanced phagocytosis function of mononuclear macrophage (P<0.01). And the all of the immune items of compound polysaccharides were notably higher than those in M3000 group. The result indicated that the immune-enhancing activity of low molecular polymannuronic acid was enhanced by combining with polysaccharides from Ganoderma Lucidum. Although there is no significant difference between the two groups, all of the immune items of compound polysaccharides group were higher than those in GLU-1 group. All the result showed that, the compound polysaccharides had immune-enhancing effects. And this research offered definite theory for the further exploitation of immune-enhancement medicine or health food.
