| Objective To observe the therapeutic effect of the Different Combination of Compound Lizards Powder (DCCLP) on Chronic Atrophic Gastritis (CAG), and explore the mechanism of the therapeutic action of DCCLP on gastric mucosal injure.Methods 90 SD healthy male rats were randomly divided into normal control group,model control group,Vitamycin treated group,compound powder lizards 80 mesh treated group,compound powder lizards 100 mesh treated group,DCCLP (80 mesh+100 mesh) treated group.Except normal control group rats,others were made to model of CAG with 2% salicylic acid and 55℃hot water which contained 150g/L NaCl and 20mmol/L sodium deoxycholate,matching irregular diet.When the model was established successfully,treated group rats were given different dose medicines(the different mesh of compound Lizards Powder or Vitamycin),after 4 weeks,all the rats were sacrificed. Their blood serum was collect to detect PGE2 by ELISA and NOS by chemic method,and their gastric mucosa was collect to detect ghrelin expression and detect the histological mucosa changes by HE.Results The PGE2 levels of treated groups were significantly higher than the model group (P <0.01), while the NOS levels were lower (P <0.01); The PGE2 level of the DCCLP group was higher than 80 mesh and 100 mesh group (P<0.01), while the NOS was lower (P<0.01).The mucosal pathology of treated groups has significant reversing. By image analysis system IHC measurement of optical density, ghrelin expression of treated groups were significantly higher than the model group (P<0.01); ghrelin expression of the DCCLP group was higher than 80 mesh and 100 mesh group (P<0.05).Conclusion The conspicuous therapeutic action of DCCLP on CAG, the quantity of PGE2 in serum was higher, serum NOS level was lower and the gastric mucosal expression of ghrelin was increased.By improving the barrier functions of gastric mucosal, increasing the role of gastric motility, etc, the protection of gastric mucosal will be achieved. |
