Effects Of The Different Combination Of Compound Lizards Powder On The Expression Of Proteins Of Stat3Pathway With Precancerous Lesions Of Gastric Cancer In Rats

Objective:To study the effects of the different combination of compoundlizards powder on the expression of Stat3,Bcl-2,Bcl-xl and Mcl-1proteins of Stat3pathway with precancerous lesions of gastric cancer in rats,to investigate itsfunctional mechanism of TCLP on PLGC in molecular level.Methods:120male SD rats were randomly divided into blank control g-roup(A),TCLP80mesh treatment group(B),TCLP100mesh treatment group(C) and TCLP equal mixing treatment group with80mesh and100mesh(D),the model control group with precancerous lesions of gastric cancer(E),Vitacoenzyme treatment group(F),except for A group,other groups make themodel of PLGC with N-methyl-N′-nitro-N-nitrosoguanidine,irregular di-et and emotional stimulation factor for8weeks.Respectively use TCLP80mesh,TCLP100mesh,TCLP equal mixing treatment group with80mesh a-nd100mesh paste suspension,physiological saline and Vitacoenzyme pastesuspension treated for12weeks after modeling success,then observe the ga-stric mucosa pathological changes of rats and detect the Stat3,Bcl-2,Bcl-xland Mcl-1proteins’ expression of gastric tissue with immunohistochemicalmethod.Results:1.The rats’ gastric mucosa pathological results:The TCLP treatmentgroups were repaired more apparently,the atrophy partially dissipated,intestinalmetaplasia had been significantly improved,dysplasia to normal,D group had the best effect.Compared with TCLP treatment groups,F group had less effect in treatthe impaired mucosa,E group had little change.2.Through the analysis of the Image-Pro Plus6.0system on immunohi-stochemical image measurement of optical density,compared with E group,o-ther groups had significance difference(P＜0.01) in the Stat3,Bcl-2,Bcl-xl a-nd Mcl-1proteins positive expression.TCLP treatment groups were lower t-han F group(P＜0.05)in the Stat3,Bcl-2,Bcl-xl and Mcl-1proteins positive e-xpression,in which D group had the most significant difference(P＜0.01).A-mong TCLP treatment groups,80mesh and100mesh had no statistical sign-ificance(P＞0.05),but D group there was differences in comparison(P＜0.05).Conclusion:1.TCLP can partially reverse the gastric mucosa gland atrophy,improve intestinal metaplasia,reduces epithelial hyperplasia,promotes dysplasiaregression and abnormal cells normal differentiation and mature,has the effect ofreverse PLGC.2.TCLP can reduce the expression of Stat3,Bcl-2,Bcl-xl and Mcl-1proteins.
