| Objective:To investigate the protective effects and mechanism of baicalin (Baicalin, BA) on renal ischemia-reperfusion injury.Methods:1 Experimental animals and groups: 24 healthy male Sprague-Dawley rats(weight (260~280g), were randomily divided into four groups: sham group(n=6), Control group(n=6), baicalin pretreatment low-dose group (Low group,n=6),and high-dose group (High group,n=6).2 Experimental model: (1)Sham group: right nephrotomy was performed, without clipping left renal artery. (2)Control group: After right nephrotomy, the left renal artery was clamped for 60min. (3) baicalin pretreatment groups: After right nephrotomy, baicalin solution was given via vena cava, then left renal artery was clamped for 60min, blood sample(1.5-2ml) were taken via the inferior vena cava after 2 hours and 6 hours of I/R, then rats were scraficed and kidney tissue specimens were taken.3 Drugs given: Sham group and Control group were injected saline solution(1.5ml) via the inferior vena cava. Baicalin pretreatment groups were given the same volume baicalin (low-dose group and high dose group, respectively).4 Measure indexs: Blood samples: serum Cr was measured by picric acid method ; serum MDA was measured by thiobarbituric acid(TBA); serum SOD was measured by xanthine-oxidation(XOD). Renal tissues: renal swelling degree, H-E staining of renal tissue for pathological observation, immunohistochemical determination of NF-κB, TUNEL assay apoptosis of renal tubular cells were performed . Results:1 Renal ischemia reperfusion injury (IRI) after baicalin on renal function and biochemical effects: Control group and baicalin pretreatment group than in the Sham group showed a increasing trend of serum Cr (P <0.05and P <0.01, respectively), Scutellaria pretreatment group than in the Control group glycosides Cr decreased significantly (P <0.01, P <0.01). Baicalin pretreatment low dose high dose group was significantly lower Cr levels (P <0.01). Serum MDA levels: Control group and baicalin pretreatment group than in the Sham group MDA levels were significantly higher (P <0.01, P <0.01), baicalin pretreatment group than in the Control group was significantly decreased (P <0.01); baicalin pretreatment low dose high dose group MDA levels were significantly decreased (P <0.01). Serum SOD levels: Control group and baicalin pretreatment serum SOD levels than the Sham group showed a certain degree, respectively, decreased (P <0.01, P<0.01). Baicalin pretreatment group than in the Control group showed significantly higher levels of serum SOD trend (P <0. 01), baicalin high dose group with the lower dose group, SOD levels were significantly increased (P <0. 01).2 Baicalin on the renal pathology of post-IRI: Control group and baicalin pretreatment group showed a certain degree of pathological changes (tubular epithelial cell swelling, vacuolar degeneration, etc.), baicalin pretreatment group than in the Control group Pathological changes was reduced. Control group in the IRI kidney tissue water content than after the Sham group, the high baicalin pretreatment group (P <0.01); baicalin pretreatment renal tissue water content lower than the Control group (P <0.01). High baicalin pretreatment group compared with the Low group was significant difference (P <0.01).3 Baicalin on renal IR after renal inflammation the expression of factors: with the Sham group, Control group and baicalin pretreatment NF-κB expression with varying degrees of upward trend (P <0.01, P <0.01), Scutellaria Pretreatment group than in the Control group glycosides NF-κB expression was significantly decreased (P <0.01), high-dose group baicalin NF-κB low dose group was significantly decreased (P <0. 01). TUNEL positive cells (per high power field): baicalin pretreatment group was significantly lower than the Control group (P <0.01). Baicalin pretreatment high dose group was significantly lower than low-dose pretreatment group.Conclusion:1 After renal ischemia-reperfusion(RIR), serum Cr and MDA increased, on the contrary, serum SOD decreased. Tubular cell damage, the expression of NF-κB increased2 Baicalin can reduce the serum Cr, MDA, the expression of kidney tissue NF-κB, and increase serum SOD levels, indicating that baicalin has significant protective effect on ischemia-reperfusion injury. The mechanism may be related to its increased activity of the antioxidant enzyme activity, reduced lipid peroxidation, inflammation and apoptosis.3 Baicalin can reduce renal tisue swelling after I/R.4 Baicalin on renal ischemia-reperfusion injury was being some positive correlation. |
