| Objective:With the guide of traditional Chinese medicine theory and modern medical theory, this experiment explores Zexie Tang lipid-lowering mechanism by observing the influence which the different concentrations Zexie Tang act on experimental hyperlipidemia rats after we establish a stable, convenient hyperlipidemia rats model, and provides scientific basis for lipid-lowering and reference for clinical medicine.Methods:60 only healthy SPF male Wistar rats are assighed randomly into six groups of 10, respectively is:the normal group (the normal control group), the model group (the hyperlipidemia model control group), the positive group (the simvastatin group), the ZXT1 group (the low dosage Jin Kui Zexie Tang group), the ZXT2 group (the middle dosage Jin Kui Zexie Tang group) and the ZXT3 group (the Zexie Tang high dosage group). These just bought rats are fed 7 days for adaptation, the experimental process reference laboratory animal protection and use guide. Then, the normal healthy rats are given ordinary feed, while the rats of the rest groups are given a high-fat feed for four continuous weeks. Then the normal group and the hyperlipidemia model group rats are given distilled water 20ml·kg-1·d-1 by Irrigating gastric (ig), the positive group rats are given 7.2mg·kg-1·d-1by Irrigating gastric (ig), the ZXT1 group, the ZXT2 group and the ZXT3 group rats are given Jin Kui Zexie Tang the original medicinal materials 3.78g·kg-1·d-1,11.34 g·kg-1·d-1 and 37.8 g·kg-1·d-1 by Irrigating gastric (ig). At the same time, each group rats kept the original feed. The progress will take 4 weeks. Each group rats urine is collected by the end of the experiments, then, these rats are fast and given water for 24 hours, and taked the artery blood from left or right common carotid artery after anesthesia by administrating narcotic drugs, these blood was used for biochemical indicators measure, the rats liver, spleen and kidney are removed, part of the livers are used for pathological microscopic observation.Results:Compared with the normal group, the hyperlipidemia model rats serum TC, LDL-C, TG, ALT, AST, NEFA, GLU and BUN have significantly increased (P< 0.01), liver viscera coefficient have significantly increased (P< 0.05), HDL-C have significantly reduced (P< 0.01), urine volume, SOD, MDA, Cr, spleen and kidney viscera coefficient have no significant differences. Compared with the model group, the positive group rats TC, LDL-C, TG, BUN in the serum, hepatic CYP7A1 mRNA level have reduced significantly (P< 0.01), hepatic HMG-CoA mRNA level have a significant reduction(P< 0.05), serum HDL-C have a significantly increased (P< 0.01), liver, spleen and kidney visceral coefficient, hepatic LDL-R gene expression level, urine volume, SOD, MDA, ALT, AST, Cr, NEFA, GLU have no significant differences; the ZXT1 group rats serum TC, LDL-C, TG have significantly reduced (P< 0.01), hepatic HMG-CoA mRNA, CYP7A1 mRNA level and serum BUN have a significant reduction (P< 0.05), serum HDL-C have a significantly increased (P< 0.01), urine volume have significantly increased (P< 0.05), liver, spleen and kidney visceral coefficient, hepatic LDL-R mRNA level, SOD, MDA, ALT, AST, Cr, NEFA, GLU have no significant differences; the ZXT2 group rats, serum TC, LDL-C, TG, BUN have significantly reduced (P< 0.01), hepatic HMG-CoA mRNA and CYP7A1mRNA level, and serum NEFA have a significant reduction (P< 0.05), serum HDL-C have a significantly increased (P< 0.01), hepatic LDL-RmRNA level have significantly increased (P< 0.05), liver, spleen and kidney viscera coefficient, urine volume, SOD, MDA, ALT, AST, Cr, GLU have no significant differences; the ZXT3 group rats serum TC, LDL-C, TG, BUN, liver CYP7A1 mRNA and the HMGmRNA level have significantly reduced (P< 0.01), serum Cr have a significant reduction (P< 0.05), serum HDL-C have a significantly increased (P< 0.01), liver, spleen and kidney visceral coefficient, hepatic LDL-R mRNA level, urine volume, SOD, MDA, ALT, AST, NEFA, GLU have no significant differences.Conclusion:1 this experimental hyperlipidemia model is successful.2 Jin Kui Zexie Tang have a lipid-lowering effect.3 Jin Kui Zexie Tang achieve lipid-lowering by inhibiting the cholesterol absorption, or reducing the hepatic HMG-CoAmRNA level and enhancing the hepatic LDL-R mRNA level.4 high blood lipid and high blood glucose have a close relationship. |
