Objective:To optimize the extracting process of of ethanol extracts of Rhizoma Alismatis and Rhizoma Smilacis Glabrae and observe their hypouricuria effects.Methods:Orthogonal test design was utilized to optimize the extraction conditions including the concentration of ethanol, extraction duration per time, extraction times and amount of ethanol when alisol B23-acetate and astilbin were selected as the evaluation index for Rhizoma Alismatis and Rhizoma Smilacis Glabrae respectively. Mice model of hyperuricemia were established by the subcutaneous injection of oxonic acid potassium and intragastric administration of hypoxanthine. The concentration levels of alisol B23-acetate, astilbin and serum acid were determined by HPLC. Results:Extraction times had significant effect on the content of alisol B23-acetate extracted from Rhizoma Alismatis. Extraction duration per time, extraction times and amount of ethanol had significant effects on the content of astilbin extracted from Rhizoma Smilacis Glabrae. The serum uric acid levels of the model group, the Rhizoma Alismatis group, the Rhizoma Smilacis:Glabrae group, the combination of Rhizoma Alismatis and Rhizoma Smilacis Glabrae group were 79.69±68.64,14.47±3.75,20.46±12.00,13.30±3.34μg-ml-1 respectively.Conclusions:The optimum extraction process of Rhizoma Alismatis was to extract two times using sevenfold 70%ethanol and 1 h per time. The optimum extraction process of Rhizoma Smilacis Glabrae was to extract two times using sevenfold 90% ethanol and 2 h per time. Both of Rhizoma Alismatis and Rhizoma Smilacis Glabrae suggested considerable hypouricuria effects and indicate no significant synergistic effect between them.
