Objective:The study was to explore the effect and the possible mechanism of PNS treating pulmonary contusion induced by heavy free-fallers on rabbits by measuring tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β) and malondialdehyde (MDA), combined with wet to dry proportion and pathological changes of lung tissues, providing the experimental and theoretical evidence for the clinical using of PNS.Methods:24 healthy adult rabbits were randomly divided into control group, contusion group and treatment group(n=8). Control and contusion groups were pumped into the saline (5 ml/kg) after moulding five minutes by the right ear margin vein. The treatment group were pumped the injectable venous thrombus tong (freeze-dried 150 mg/kg), all were pumped in within ten minutes. The blood samples were collected from left femoral vein 0.5h before impacting and 0.5h,2h,4h,6h, after impacting. At the end of the experiment, the serum was used to detected TNF-a, IL-1βlevels, small lung tissue were collected to measured Wet/Dry weight ratios (W/D) and MDA content, pathological changes of lung tissue were observed in the light microscope.Results:(1) Compared to the control group, serum TNF-αlevel in contusion group began to increase after 0.5 h of contusion, reaching to peak at 2 h, close to the control level at 4~6 h, (P<0.01); serum TNF-αlevel in treatment group increased more slowly, higher than control group after 2 h, but significantly lower than the contusion group (P<0.05), near to the control level after 4~6 h. (2) Serum IL-1βlevel in contusion group began to increase after 0.5 h of contusion, reaching to peak at 2 h, decreased significantly at 4~6 h, (P<0. 01); serum IL-1βlevel in treatment group showed slow declining, compared with contusion group (P<0.01); but still significantly higher control group (P<0.01). (3) The MDA content of lung tissue in contusion group was significantly higher than the control group (P<0.05), which was decreased obviously after PNS intervention (P<0.05); W/D in contusion group was higher than control group (P<0.05), which was also reduced in treatment group (P<0.05). (4) Pathological studies showed increased lung contusion volume, scattered patches in the congestion, sliced red solid-like change; microscope showed pulmonary interstitial edema, exudation, hemorrhage, granulocyte infiltration and destruction of alveolar structure. Pathological injury was significantly reduced in treatment group compared with contusion group.Conclusion:PNS can reduce lung tissue damage induced by inflammation and oxidative stress by lowering serum TNF-α, IL-1βlevels and MDA levels of lung tissue; PNS improves edema and neutrophil infiltration; PNS may have better protection function for the rabbit acute pulmonary contusion.
