Experimental Study On Astragalus Decoction Treatment Of Type 2 Diabetes Mellitus

Objectives To observe the therapeutic effect of Astragalus Decoction on rats with type 2 diabetic mellitus and primarily explore its possible mechanisms for clinical treatment.Methods Select 90 male Wistar rats of Specefic Pathogen Free,weighted 140±10g.All the experimental animals were fed under the SPF environment for a week,and then select 10 rats as the normal control group;the other rats were fed by high fat diet for 4 weeks,and then the rats were acquired through intraperitoneal injection of STZ of 35mg?kg-1 to establish the diabetic rat model.The diabetic rats were randomly divided into 6groups:model control group, Western medicine group (Merformin Hydrochloride group), Chinese medicine group(ShenqiJiangtang Powder group ),the low dosage of Astragalus Decoction group, the middle dosage of Astragalus Decoction group and the high dosage of Astragalus Decoction group,10 rats each.All the medicine were given by ig in the rats of treatment group once a day for 8 weeks,and the same volume 0.9% saline were given in the normal control group and model control group. Record the condition of monitoring blood glucose and weight of experimental animals every week. At the end of the experiment, after the anesthesia of 10% chloralic hydras ,the blood was collected from the abdominal aorta to examine FBG,TC,TG,HDL-C,LDL-C,Ins and HbA1c.The pancreas of rats were fixed in the fixative solution of paraformaldehyde for morphological and immunohistochemical test.The experimental data were deal with SPSS17.0 software and then observe the changes before and after treatment.Results Rats model of type 2 diabetes can be established by high fat diet fed and injected intraperitioneally with low dosage of STZ .The symptoms of polydipsia, polyphagia, polyuria and emaciation can be reduced by Astragalus Decoction.When the experiment finished, compare with the normal control group, body weights of rats model of type 2 diabetes obviously decreased (P<0.05),but compare with the model control group, body weights of treatment groups increased (P<0.05).The level of FBG of model rats descends in the treatment groups after the experiment(P<0.05).Compare with the middle and low dosage of Astragalus Decoction group,the hypoglycemic effect of the high dosage of Astragalus Decoction group is much better(P<0.05).No difference among Western medicine group, Chinese medicine group and the high dosage of Astragalus Decoction group(P>0.05).Compare with the model control group,the content of TC,TG,LDL-C and HbA1c in each Astragalus Decoction group decrease(P<0.05),HDL-C and Ins increase( P<0.05 ) .Hematoxylin-eosin staining and Immunohistochemical staining:compare with the model control group,the number of pancreatic isletβcells of treatment groups increase, and the structure ofβcells relatively normal,but those changes in high dosage of Astragalus Decoction group were more significantly.Conclusions Astragalus Decoction could reduce the FBG and regulate the lipid metabolism of rats with type 2 diabetic mellitus which maybe achieved through promote Ins secretion,improve insulin resistance and protect the pancreatic isletβcells.
