Objective Study on relationship between changes of AH antigen in part of AML and MDS patients and ABO gene promoter CpG island methylation.Methods Adopt flow cytometry technique to test A/H antigen expression and use Methylation-specific PCR to test ABO gene promoter CpG island whether or not methylated.Results There is evident significance in A antigen positive expression rate and Mean fluorescence intensity of AML /MDS patients compared with normal control(P <0.05),while there is no significance in H antigen positive expression rate and Mean fluorescence intensity (P>0.05).14/25(56%) cases of AML/MDS showed ABO gene promoter CpG island methylation . 8 cases of 16 who showed that reduction of A antigen positive expression rate and(or) Mean fluorescence intensity indicated ABO gene promoter CpG island methylation(4 AML, 4 MDS), and the other 8 were not methylated(4 AML, 4 MDS). Otherwise, 6 of 9 who did not decrease of A antigen positive expression rate and(or) Mean fluorescence intensity turned out methylation(3 AML, 3 MDS).Conclusions Blocking of process of H antigen converted into A antigen leads to reduction of A antigen expression with AML/MDS, and parts of the patients show ABO gene promoter CpG island methylation ,but whether it is typically related with ABO gene promotor CpG island methylation ,we need more samples to explore.
