| Objective To study the association between the polymorphism of site rs324015in STAT6(signal transducers and activators of transcription 6)gene and the genetic susceptibility to asthma in children of Qingdao Han ethnic group. Methods This was a case control study. PCR—RFI technology was applied to investigate the polymorphism of site rs324015(A/G)in STAT6 gene among 113 asthmatic subjects and 87 unrelated healthy han children, and Hardy—Weinberg hereditary balance test was used to examine the coincidence of gene distribution.Results (1)The distribution of genotype frequencies of site rs324015 accorded with Hardy-Weinberg's equation law, being colony representative. (2) the frequencies of STAT6 GG, GA and AA genotypes of the SNP at locus rs324015 were 15.9%,46.9% and 37.2% in asthmatic group, respectively They were 21.8%,59.8% and 18.4% in normal control group, respectively.In comparision with GA and GG genotypes carrier,the risk of asthma increased in those with A A genotypes(OR(AA/GA)=2.575,95% CI:1.290-5.141,p<0.01; OR(AA/GG)=2.771,95 %CI:1.167~6.577, P<0.05).The frequencies of the two alleles G and A were 39.38% and 60.62% in asthmatic group.And 51.72% and 48.28% in normal control group, respectively.Prevalence of asthma in alle A carrier was as much 1.69 times as that of those without alle A[OR (A/G)= 1.649,95%CI:1.106~2.459, P<O.05]. There was significant difference in the frequency distribution of genotype and allelic genes of site rs324015 in STAT6 gene between two groups(X2=8.445, P<0.05; X2=6.058, P<0.05) Conclusion the polymorphism of site rs324015 in STAT6 gene may contribute to disease susceptibility in asthma, the risk of asthma increases in carrier with mutant allele A,and allele A may be superior gene in prevalence of asthma. |
