Objective:To study the distribution frequency of single nucleotide polymorphism(SNP)of the interleukin-10 gene promoter rs1800896 and rs1800871 loci in Guangxi Black Zhuang children,and to compare the distribution of genotype and allele frequency with different ethnic and geographical populations.To compare the differences in genotype and allele frequency distribution with different ethnic groups and regions,and to preliminarily analyze the correlation between single nucleotide gene polymorphism(SNP)at the promoters of interleukin-10 gene rs1800896 and rs1800871 loci and bronchial asthma in Guangxi Black Zhuang children.Methods:100 cases of Guangxi Black Zhuang children with bronchial asthma and 100cases of children with healthy physical examination were collected as study subjects,and the ligase detection reaction(LDR)technique was applied to genotype the interleukin-10 gene promoter rs1800896 and rs1800871 loci to test and verify the population representation of the samples was verified by Hardy-Weinberg equilibrium at each locus.The genotypes of the asthma and healthy groups were calculated using the gene-counting method,as well as the frequencies of the allelic genotypes of the asthma and healthy groups.The chi-squared test was used to compare differences in the distribution of single nucleotide polymorphisms by sex among healthy children of the Black Zhuang ethnic group in Guangxi,and to compare differences in the distribution of genotypes and allele frequencies with other ethnic and geographic populations.The linkage disequilibrium and haplotype analysis was done using the SHEsis online tool.Count data were analyzed using chi-square test,and measurement data were expressed as(mean±standard deviation),and independent samples t-test was used for comparison between groups.Statistical analysis of the data was performed using SPSS22.0software,test level:α=0.05,and differences were considered statistically significant at P<0.05.Results:(1)The interleukin-10 gene promoter rs1800896 locus detected CC,CT and TT genotypes in the healthy group of Guangxi Black Zhuang children,and the genotype distribution frequencies were CC1.0%,CT14.0%,TT85.0%;The frequencies of the alleles are C8.0%,T92.0%;The interleukin-10 gene promoter rs1800871 locus was detected in the healthy group of Guangxi Black Zhuang children with genotype distribution frequencies A43.0%,AG48.0%,GG9.0%;The frequencies of the A67.0,G33.0alle distribution;(2)Interleukin-10 gene promoters rs1800896 and rs1800871 loci genotypes and alleles were not statistically significant between the sexes(P>0.05);(3)the genotype and allele frequencies of the interleukin-10 gene promoter locus rs1800896 in children of the Guangxi Black Zhuang Health Group were similar to those published in the data of the 1000 Genomes Project(1000Genomes)for African Nigerians(ESN),the(YRI)in Ibadan,Nigeria,Africa,Iberians(IBS)in Spain,and Gujarati Indians(GIH)in Houston,USA;the differences in genotype and allele frequencies were statistically significant(P<0.05)when compared with those of Han Chinese(CHS)in southern China;the differences were not statistically significant(P>0.05)in Guangxi Black Zhuang the genotypes and allele frequencies of the interleukin-10 locus rs1800871 in healthy group children were comparable to those published in the data of the1000 Genomes Project(1000Genomes)for African Nigerians(ESN),the Ruba of Ibadan,Nigeria(YRI),the Iberians of Spain(IBS),and the Gujarati Indians of Houston,USA(GIH).the differences in genotype and allele frequency comparisons were statistically significant(P<0.05);there were no statistically significant differences in genotype and allele frequency comparisons with the Han Chinese of Southern China(CHS)(P>0.05);(4)there were no statistically significant differences in gender and age between children in the asthma and healthy groups(P>0.05);(5)the selected samples were all consistent with Hardy-Weinberg’s law of equilibrium and were representative of the group(P>0.05);(6)the results of linkage disequilibrium analysis showed that rs1800896 and rs1800871(D,=1,r~2=0.146),a group of loci with strong linkage disequilibrium;(7)haplotype frequencies showed that the present study used the SHEsis online tool for the interleukin-10 gene promoter rs1800896 and rs1800871 loci for haplotype typing,and it was found that C-G,T-A and T-G haplotypes existed in both asthma and control groups,among which T-A haplotype was the most common,and haplotypes accounted for 68.5%and 67.0%in the asthma and control groups,respectively.Conclusion:1.The polymorphisms exist at the loci rs1800896 and rs1800871 of the interleukin-10 gene promoter in children from the Guangxi Black Zhuang healthy group;2.The polymorphisms present at loci rs1800896 and rs1800871 of the interleukin-10 gene promoter in children from the healthy group of Guangxi Black Zhuang have a regional character;3.There was no correlation between the genotype of the interleukin-10 gene promoter rs1800896 and rs1800871 and the frequency of their allele distribution and asthma risk;4.Haplotyping of the interleukin-10 gene promoter rs1800896 and rs1800871 in children with bronchopulmonary asthma in Guangxi Black Zhuang revealed C-G,T-A and T-G haplotypes in both asthma and control groups,with the T-A haplotype being the most common;5.Strong linkage disequilibrium exists between the interleukin-10 gene promoter rs1800896 and rs1800871 in children with bronchopulmonary asthma in Guangxi Black Zhuang.
