Objective Discussing the relation between SNP in the BRCA1 gene promoter area rs11655505, rs73625095, rs78350562 and sporadic breast cancer predisposition. Methods The ASA-PCR and DNA sequencing methods were used to analyze the SNPs of BRCA1 gene promoter region in 200 patients with breast cancer (all confirmed diagnosis) and equal quantity of nomal women, and the role of SNPs of BRCA1 gene promoter region was dicussed. Results The satistics showed the A/G genotype frequencies of BRCA1 gene promoter region rs11655505, rs73625095, rs78350562 of patients with breast cancer (75%,68%,48.5%) were separately higher than those of the nomal person (40%,15%,16%); The A/A genotype frequencies of BRCA1 gene promoter region rsl 1655505 (7%) was separately lower than those of the nomal person (30%); The G/G genotype frequencies of BRCA1 gene promoter region three sites of patients with breast cancer (18%,32%,51%) were separately lower than those of the nomal person (30%,84%,83%); There was notable difference in the A or G alleles in rs73625095, rs78350562 site between the cases and the controls (χ2=35.368, P=0.000); Further analysis showed that the relation between A/G genotype frequency in three sites of breast cancer group BRCA1 gene promoter region and lymph node metastasis or not is inversely proportional. Conclusion The A/G genotypes in BRCA1 gene rs1165505, rs73625095, rs78350562 site may have close relation with the happen of sporadic breast cancer and translation of lymph node metastasis. The alleles in rs73625095, rs78350562 sites may be genetic risk factors of sporadic breast cancer.
