Objective:To explore the relationship between BRCA1 rs1060915 gene polymorphism and genetic susceptibility to Luminal B breast cancer,and to analyze the correlation between different BRCA1 rs1060915 genotypes and clinicopathological features of Luminal B breast cancer.Methods:The peripheral blood samples of 511 patients with Luminal B breast cancer with complete clinicopathological data were collected as the study group and 605 healthy women as the control group.The genomic DNA,of the whole blood was extracted and the DNA template containing the SNP site region was amplified by PCR,and then specific extension primers were used for single base extension reaction with PCR products.The base difference caused by SNP polymorphism was detected by mass spectrometry gene analyzer,and the SNP locus was genotyped by molecular weight difference.The data of this study were analyzed by SPSS24.0 and online SNPStats program(https://www.snpstats.net/start.htm)).The quantitative data were statistically described by mean ±standard deviation,and the qualitative data were described by constituent ratio.T-test was used to compare the age between the case group and the control group,and the goodness-of-fit test of Hardy-Weinberg equilibrium(HWE)test was used.After adjusting the age,multivariate Logistic regression analysis was used to analyze the association between rs1060915 loci and breast cancer.The optimal genetic model was selected by AIC criterion,and the correlation between genotypes and clinicopathological features of breast cancer was analyzed by means of interval 2 test.The difference was statistically significant(P < 0.05).Results:By comparing the distribution of BRCA1rs1060915 genotypes in the study group and the control group,we found that there was no significant difference in allele distribution between the case group and the control group.There was no significant difference in genotype distribution between case group and control group under dominant model and recessive model(P > 0.05),but there was significant difference between co-dominant model and overdominant model(P < 0.05).It is suggested that the genotype of rs1060915 locus may be related to the susceptibility to breast cancer.Under the optimal genetic model,the distribution of clinicopathological characteristics of breast cancer was different among different genotypes.The results showed that the main pathological type of patients with GA genotype was invasive ductal carcinoma.There was no significant difference between other clinicopathological features and genotypes(P > 0.05).Conclusion:BRCA1 rs1060915 GA genotype is associated with susceptibility to Luminal B breast cancer.The main pathological type of Luminal B breast cancer with BRCA1rs1060915 GA genotype was invasive ductal carcinoma.
