Lung cancer is the most common malignant tumors in the world, the incidenceand mortality of the malignant tumors disease is in the first place. The incidence of non-small cell lung cancer amonts to80%, and the curing rate is below15%.Although the technology of diagnosis and treatment has been improved,the5an existence rate has not improved obviously. The main reasons is that the lack inter-vention of the occurrence,development early diagnosis to tumors. The tumor ce-llsmainly live in a relative lack of oxygen environment. The wa-rburg effect of tumorcells would produce much acidity material such as lactic acid and pyroracemic acid. But if the sinotrans of the lactic acid can't accordwith generation, lactic acid will accumulat in the cells, that will cause the pH value fall down within the cell, and make the cancer cells acidification and apoptosis. If keeping the state in cells glycolysis, lactic acid must be transported out of the cell. Monocarboxylate transporters plays a very important role in discharge lactic acid. Among them MCT1as a transport through membrane protein exists in many entity tumor cells, such as breast cancer, ovarian cancer, colorectal cancer and so on. MCT1must depend on the extracellularmatrix metalloproteinases inducer(CD147) and CD44in the cell membrane, which will make MCT1express correctly on the cell membrane site, and make lactic acid channels across the cell membrane and perform transport function. But there were no reports of detailed study about the relationship among MCT1, CD147,CD44in non-small cell lung cancer. Therefor, the passage of MCT1,CD147, CD44will become a new targets to control tumor growth and lymph node metastases. It may become the detection of malignant degree and the lymph node metastases and the important assessment index. There is no doubt that it will open a new road for the treatment of lung cancer.Purpose The tumor cells suiting to anoxia environment is the important biological characteristics. In recent years, the studies found thatMonocarboxylate transporters (MCTs) are important cellular pH regulators in cancer cells, especially Monocarboxylate transporters one(MCT1). MCT1exists in the cell membrane surface with CD147and CD44, which mainly Makes the tumor cells adapt to the hypoxic conditions。However, the expression of relevance and significance of MCT1, CD147,CD44protein in non-small cell lung cancer is still poorly understood. In the present study, we analysed MCT1,CD147,CD44protein expression in non-small cell lung cancer, and their clinico-pathological value.MethodsFifty-six specimens of the NSCLC, peritumoral tissues as experimental group and twenty pulmonary benign diseases as control group were studied. The expression of MCT1,CD147,CD44mRNA and protein in specimens of those tissues were studied by real-time quantitative reverse transcriptase polymerase chain reaction (real-time quantitative RT-PCR) and Immunohistochemistry.Result1. Real-time quantitative reverse transcriptase polymerase chain reaction (real-time quantitative RT-PCR)①he expressions of MCT1,CD147,CD44mRNA in specimens of NSCLC were significantly higher than those of the peritumoral and pulmonary benign diseases tissues (P<0.05);②Significantly correlations among MCT1,CD147,CD44(t=2.398P=0.020, t=8.299P=0.000, t=5.712P=0.000);③The expressions of MCT1,CD147,CD44mRNA in NSCLC were not correlated with age, gender, site and dimension of lesion, and pTNM stage, but correlated significantly with and types of histological (t=2.214P=0.031, t=2.242P=0.021, t=2.126P=0.027) and lymph node metastasis(t=2.075P=0.043, t=2.166P=0.036, t=5.125P=0.020). In addition, MCT1and CD44is correlated significantly with degree of differentiation (t=2.450P=0.017, t=2.546P=0.021).2. Immunohistochemistry①he positive expressions rate of MCT1,CD1477, CD44protein were significantly higher than those of the peritumoral and pulmonary benign diseases tissues (P<0.05);②Significantly correlations between MCT1(in the cell membrane surface) and CD147,CD44, respectively is (x2=7.284, P=0.007; X2=6.522, P=0.011), MCT1(in intercellular substance) is not correlated with CD147(x2=1.601, P=0.206,CD44(x2=1.299, P=0.254);③The expressions of MCT1,CD147,CD44protein in NSCLC were not correlated with age, gender, site and dimension of lesion, and pTNM stage, but correlated significantly with types of histological (x2=18.939P=0.000, x2=3.997P=0.046, x2=3.982P=0.046, x2=4.235P=0.040) and lymph node metastasis (x2=4.806P=0.028, x2=4.806P=0.028, X2=7.748P=0.005, x2=12.493P=0.000), in addition, MCT1(in the intercellular substance)is correlated significantly with degree of differentiation (x2=7.153P=0.028, x2=5.486P=0.019) and smoking (x2=6.623P=0.010).ConclusionThe expressions of MCT1,CD147,CD44mRNA and protein in specimens of NSCLC are significantly higher than those of the peritumoral and pulmonary benign diseases tissues, all the data reveals that they are significantly correlated with each other. And expressions higher in squamous carcinoma than in adenomatous carcinoma.MCT1,CD147,CD44may participate in the occurrence and development of cancer. So they may act as the test NSCLC and prognosis index.
