Centipede alga(Grateloupia filicina), a kind of Marine red algae, whichbelongs to Rhodophyta, Florideae, Cryptonemiales, Halymeniaceae, is Widelydistributed all over the coastal warm temperate zone in the world. Polysaccharide, theimportant main component in centipede alga, has shown several biological activefunctions such as antiangiogenic, antivirus, Antitumor, anticoagulant, anti-thrombotic,anti-fatigue and hypoxia. At present the relevant research report about centipede algais relatively sparse, the reducing blood fat and lowering blood sugar active researchon centipede alga has not yet been reported interiorly.This paper took Grateloupia filicina from Fujian Fuqing sea areas as rawmaterial, discussed the extraction and purification process of Grateloupia filicinapolysaccharide, determined the basic physical and chemical properties of centipedealgae, and studied the antioxidant, antiatheroscloresis and antidiabetics activity ofGrateloupia filicina through the animal study. The main research work andconclusions were as follows:(1) According to analysis of polysaccharide yield index of Grateloupiafilicina by comparing four extraction methods such as the water extraction, alkaliextraction, enzymatic hydrolysis and microwave-assisted extraction,the experimentdetermined to extract centipede algae polysaccharide with hot water formulation.Hot water extraction and alcohol sink centipede algae polysaccharide, tookextraction temperature, extraction time and solid-liquid ratio as the three factors,the optimal combination of Grateloupia filicina polysaccharide extracting processis selected by single factor experiment and the orthogonal experiment design:temperature of100℃, time5h, solid-liquid ratio1:60, The centipede algaepolysaccharide yield can reach59.42%.(2) Crude polysaccharide separation and purification: by comparing the Sevageand TCA removal protein method, TCA removal protein method had no significanteffect in this experiment, the experiment take Sevag removal protein method5timesas Grateloupia filicina polysaccharides removal protein method, using glucan gelG-150through the chromatography column for purification centipede algapolysaccharides and collect eluent. Determined the distribution of molecular weight in3.5×105~1.2×105through the test of the collection of the eluent sugar content, and showed the molecules within the scope of polysaccharides have clearance activity onhydroxyl radicals (OH·).(3) Polysaccharide biological activity research: The clearance on OH· andO2· of centipede alga polysaccharide were determined respectively, and theantioxidant, antiatheroscloresis and antidiabetics activity were studied, the resultsshowed that centipede alga Polysaccharide had good antioxidant effect in vitro, theclearance IC50to OH· was4.62mg/mL, the clearance IC50to O2·was0.62mg/mL,the centipede alga polysaccharide can increase the serum of the mice and had liverSOD, T-AOC, CAT vitality effect, and can effectively inhibits the growth of MDA inhigh blood fat mice; the centipede alga polysaccharide can significantly reduce the fatmice weight, by taking the centipede alga polysaccharide,the mice's serum and liverTC, TG, LDL-C were significantly lower compared with the normal mice and HDL-Cwas significantly increased, these showed that centipede alga polysaccharide haveprevention and reduction effect in mice's high blood fat, its effect was even betterthan XieZhiKang; The Grateloupia filicina polysaccharide can markedly reduce theblood glucose of high blood glucose mice and it also can control decreasing degree ofthe mice's weight and can improve the phenomenon of the mice such as the diabetes,bad mental state, body hair mixed and disorderly.
