Polysaccharide is one of the important components of Morinda Citrifolia L.(noni),its extraction process,monosaccharide composition,antioxidation and antitumor activity were researched at present,there is no literature to report the purification process,the same with antioxidant and antitumor activities of noni polysaccharide samples sank by different concentration ethanol.Paper firstly studied ultrasonic-assisted extraction and purification process of noni polysaccharide,then comparised the differences on monosaccharide composition and antioxidant with antitumor activity between polysaccharides that sank by different concentration ethanol,screened out polysaccharide whice is efficient activity.Paper contents mainly included the following:1.Ultrasonic-assisted extraction process of noni polysaccharide was explored and optimized by the response surface method.Results showed that the optimal extraction process were liquid-solid ratio 30.2mL/g,extraction time 38.5min,ultrasonic power 82.3W,extraction temperature59.6℃,actual yield of noni polysaccharide was(8.657±0.193)%.2.Decolorization process of noni polysaccharide was studied with macroporous adsorption resin.Static adsorption screening results of seven resins indicated that the decolorizing effect of HZ-214 resin and number one decolorization resin to noni polysaccharide were better.The continuing static adsorption kinetics experiment of two kinds of resins showed that the preferable decoloration process of HZ-214 resin were polysaccharide solution volume 20mL,pH 9,adsorption time 6h;the preferable decoloration process of number one decolorization resin were polysaccharide solution volume 30mL,pH 8,adsorption time 6h.Decolorizing effect of number one decolorization resin to noni polysaccharide was better than HZ-214 resin from the overall data,therefore it was number one decolorization resin for further dynamic adsorption.The preferable process of dynamic adsorption with number one decolorization resin were:sample volume 2BV,water elution volume3BV,adsorption time 3.5h,flow rate 6BV/h,noni polysaccharide purity from(23.07±0.16)%up to(60.33±0.21)%.3.Noni polysaccharide were sank by various concentration of ethanol(30%,50%,70%,90%),respectively obtained four polysaccharide samples namely A,B,C,D,analysising their monosaccharide composition and proportion with pre-column derivatization HPLC method,results indicated that four polysaccharide samples were composed of mannose,rhamnose,glucose,galactose and arabinose,but the molar percentage were different,mannose,rhamnose,glucose,galactose and arabinose molar percentage of A,B,C and D successively were1.33:4.24:77.22:9.83:7.38,2.58:2.28:52.20:31.40:11.54,1.32:6.45:47.00:32.91:12.31,3.18:3.39:27.23:45.50:20.70.4.Antioxidant activity of noni polysaccharide were investigated with DPPH·,OH·and O2-·three antioxidant activity evaluation systems,results revealed that the scavenging ability of four polysaccharide samples to DPPH·and O2-·were D>C>B>A,scavenging ability to OH·was C>B>A>D,thus antioxidant activity of C was higher.Then measured inhibition effect of noni polysaccharide on A549 cell by3H-TdR incoporation method,the results indicated that inhibition effect of A on A549 cells is stronger than others(P<0.01),B and D on A549cells were almostly same,C on A549 cells was weakest.
