The regulatory network of transcription factors in tapetum isimportant for the mature pollen development. Thus, based on theformer analysis of tapetal genetic pathway, we use semi-thin crosssections and in situ hybridization to show the genetic relationships with5tapetal transcription factors (DYT1-TDF1-AMS-MYB103-MS1). Additionally,TDF1which belongs to the R2R3-MYB transcription factor has beenfurther studied for its function. First, construct the transgenic mutant lineswith MYC/FLAG tag and identify the complementary phenotype.Second, establish the ChIP system to enrich the directly target genes ofTDF1. Third, Chip analysis finds that total1790genes are obviouslychanged (1252down-regulated and538up-regulated) in the tdf1flower buds. Aiming at the down-regulated genes of TDF1, wespeculate several candidate downstream genes directly regulated byTDF1with analysis of the ‘Electronic Fluorescent Pictograph’(e-fp)Browser dataset, function clustering and myb binding sites in promoters.Moreover, we express the TDF1protein in prokaryotic vector and try tomake it recovery. Finally, the yeast two-hybrid experiment is performedto study the transcriptional function region. These results will furtherindicate the regulatory pathway for the tapetum transcription factors.Meanwhile, it will provide the important clues for us to better explainhow the transcriptional regulatory pathway functions for tapetum.
