HB (Homeobox) transcription factors are one of the most important regulatoryproteins and had been reported to be involved in regulation of plant morphogenesis,hormone response, biotic or abiotic stresses. In this study, Oryza Sativa subsp.japonica was used as the material. We had cloned69HB transcription factor genesand attached them to the transcription activate expression vector (VP64-pCambia1301-UbiN) and the transcription inhibit expression vector (EAR-pCambia1301-UbiN) respectively using GatewayTMcloning technology. Then wetransferred the two kinds of vectors into rice using agrobacterium mediatedtransformation method. After we got the transgenic plants, we did several steps ofscreening including PCR and Western Blot.In this experiment, we found that OsHB40transgenic plants were of dwarfcharacteristics through field observation and statistical analysis of plant height as wellas yield data. Then we analyzed the promoter sequence of OsHB40, and it revealedthat there was a SA cis-acting element. So we used real-time quantitative PCR toanalyze the relative expression lever of OsHB40under the treatment of exogenous SA.The results showed that exogenous SA improved the expression level of HB40gene inrice leaves, suggesting that OsHB40might regulate plant height, respond to SA-induced expression, and play a regulatory role in plant development.While we analyzed the phenotype of OsHB102transgenic plants, we found thatOsHB102transgenic plants had a lower height compared with the wild-type plants. Sowe speculated that HB102might be a transcription factor that involved in plant height.After a series of prokaryotic expression and subcellular localization of HB102protein,we found that HB102was soluble protein, and may function in the cell nucleus. Yeast two-hybrid experiments found HB39(Os02g08544.1) and OFP20(Os05g36970.1) have weak interactions..
