In recent years, Marine pollution situation is worsening, Marine pollution by theformation of the "red tide" throughout the continents of the world. In our country’s wuhu,tai lake, chao lake, ect outbreak of "water China". The water eutrophication,phytoplankton multiply is red tides and water produced the main cause of China. Not onlydoes it make human in the economy suffer significant loss, and in health and even lifeunder a significant threat. Therefore, the sea water lanceolata plantation phytoplanktonreal-time effective monitoring water pollution monitoring and management as the key.This topic is the composition of the country (863) program——"Satellite remotesensing of marine technology, ocean fishing grounds", the research goal is to establishon-site determination of chlorophyll. The water chlorophyll as the water nutrition trackingindicator, rapid detection of water chlorophyll content can warning water pollution. Thepaper briefly describes the research progress and status of fluoremetry, the mechanism offluorescence, the fluorescence spectra and the principle of chlorophyll fluorescencedetermination. Preparing of the chlorophyll extract has a concentration gradient andrespectively determinating of chlorophyll content via chlorophyll-line analyzer, FL-7000fluorescence spectrophotometer and722-spectrophot-ometer at the same time. Thecorrelation coefficients of working curves obtained were0.9985,0.9799,0.9840,compared the results we know the instrument have a good linear relationship.Experimentsshow that the linear range of the maximum value of the chlorophyll-line analyzer is about440mv. But, this system is qualitative research, no quantitative calibration, just in thechange of chlorophyll concentration read relatively fluorescence intensity, it cannot getchlorophyll concentration size, but only through the fluorescence intensity of chlorophyllconcentration changes reflect the tendency of changes;Finally, the impact factors of theapparatus are discussed and some advices about how to improve the system is given.The study is of important theoretical and practical significance for establishing ofchlorophyll-site detection method. To sum up, this chlorophyll fluorescence measuringinstrument has a high sensitivity, no reagents, speed, good real-time performanceadvantages, and can be used in the lakes, rivers, and sea water on-line detection.
