As a typical tropical fruit, Jackfruit has been cultivated in China for more than athousand of years. Its flesh is delicious, often be made as dried fruit and exported all over theworld. However, other parts of Jackfruit were not used, become the production waste. TheJackfruit related research mainly concentrated in food utilization and development level, butno research take polyphenols as the object of study.In this thesis, an analytical and separation method was established to test the content ofpolyphenols in Jackfruit’s trash. At the same time, the content and the type of procyanidin wasalso been tested. At last, solvents which have different polarity was used to extract differentpolyphenol from the sample. Hope that the research in this thesis can provide references forthe development of Jackfruit’s Trash.1. By the orthogonal design based on every single factor, the Folin-CiocalteuColorimetry was optimized. The best condition was, phenol reagent:3.5mL,7.5%sodiumcarbonateon:2.0mL, color reaction time:40min. The sequence order of each factor was:sodium carbonateon> phenol reagent> color reaction time.2. From finding the optimized extraction condition, an analytical method of polyphenol’scontent in Jackfruit’s Trash was finally established: extractive time:1.5h, material-liquid rate:1:30, ethanol concentration:30%, extractive temperature:80℃. And also got the sequenceorder of each factor was: material-liquid rate> extractive time> extractive temperature>ethanol concentration. Through stability experiment, reproducibility experiment and theexperiment on the recovery rate, we knew that the sample was stable within10hours, rangewas2.6%. When n=5, RSD was1.3%. The average recovery rate was102%. From that, wecan see, this method is an accurate and reliable method. At the same time, the content ofpolyphenols in different parts of Jackfruit was greatly different: episperm had the most, thenepicarp, while endocarp hold the least.3. It was proved that there was procyanidin in Jackfruit’s trash. After the color reaction,the product was separated, determined by UV and IR, we can make sure that the monomerwas cyaniding. The procyanidin’s content in Jackfruit’s trash was0.917mg/g, occupy88.17%of polyphenols. 4. The sample was fractionated extracted by petroleum ether, ethyl acetate and butanol.The result showed that, only the ethyl acetate can purify polyphenols, extraction ofpolyphenols in22.9%out. Petroleum ether extraction efficiency is weak, can be considered inedulcoration step, removing a portion of the low polar impurities. Butanol cannot be used forextraction because of the reaction between the samples.
