The natural flavonoids and essential oil are extractive of selected plant which have wideprospect in application.In order to promote the comprehensive utilization of flowers ofEriobotrya.japonica.L, the extraction, purification, component analysis and antioxidant activitywere studied, and established production processes for the production of essential oil andflavonoids from flowers of Eriobotrya.japonica.L.In the study, flavonoids from flowers of Eriobotrya.japonica.L were obtained by the methodof ultrasound-assisted extraction. The extraction process was optimized by the Response SurfaceAnalysis method on the basis of the single-factor experiment. The optimum extraction conditionswere as follows:64%of ethanol concentration,1:44of the solid-liquid ratio,59℃of extractiontemperature,38minutes of extraction time, the flavonoids from flowers of Eriobotrya.japonica.Lextracted were106.422mg/g under these conditions. The extraction efficiency of theultrasound-assisted extraction method was higher than the traditional extraction method, lessextraction time as well. Considering the factors such as time、energy consumption and operatingconditions, the ultrasound-assisted extraction method was optimum to extract flavonoids fromflowers of Eriobotrya.japonica.L.Using macroporous resin adsorption to separate and purificate flavonoids from flowers ofEriobotrya.japonica.L through the static adsorption-desorption and dynamicadsorption-desorption experiments, the AB-8and D101resins which had better adsorption anddesorption rate were selected. Ultimately, AB-8resin was selected for subsequent purificationoptimizating experiment by static adsorption and desorption kinetics experiments. Study on theeffect of the sample solution concentration and pH, eluant concentration and pH, and the flowrate of adsorption and the flow rate of desorption for AB-8resin adsorption and desorptionperformance, the optimum adsorption and desorption conditions were that: the concentration offlavonoids3.5mg/mL, the pH value4.0, the flow rate of adsorption3BV/h,60%ethanol aseluant and the pH value6.0, and the flow rate of desorption2.5BV/h. In that case, the content offlavonoids was68.57%.The color reaction results showed that flavonoids from flowers of Eriobotrya.japonica.Lmay contained flavone, flavanone and dihydroflavonol, chalcone and isoflavones; The results ofUV-visible spectroscopy indicated that the flavonoids from flowers of Eriobotrya.japonica.L methanol solution had two adsorption peaks at283nm and326nm, respectively, which wassimilar with flavanone and dihydroflavonol spectrum, so flavonoids from flowers ofEriobotrya.japonica.L may contained flavanone and dihydroflavonol; The results of infraredspectroscopy showed that flavonoids from flowers of Eriobotrya.japonica.L had the similarspectrum with rutin, while analysis the IR spectrum adsorption peaks, we speculated thatflavonoids from flowers of Eriobotrya.japonica.L probably were acidic phenols which containedbenzene ring structure, heterocyclic carboxyl and saturated methylene, it might be flavanone anddihydroflavonol.Study of using ascorbic acid as the control, using spectrophotometry method of flavonoidsfrom flowers of Eriobotrya.japonica.L scavenging two phenyl bitter acyl radical (DPPH·),superoxide anion radical (O2-·), hydroxyl radical (·OH) to test antioxidant activity of flavonoidsfrom flowers of Eriobotrya.japonica.L in vitro. The results showed that, flavonoids from flowersof Eriobotrya.japonica.L on O2-·and·OH maximum scavenging rate all achieves80%above,whose IC50were4.5386mg/mL and2.5327mg/mL, and the scavenging effect was about to Vcstandard, which showed that flavonoids from flowers of Eriobotrya.japonica.L had strongO2-·and·OH scavenging ability; flavonoids from flowers of Eriobotrya.japonica.L onDPPH·removal capability rate was more than70%, the IC50value was0.1615mg/mL,indicating that flavonoids from flowers of Eriobotrya.japonica.L had strong DPPH·scavengingcapacity; comprehensive flavonoids from flowers of Eriobotrya.japonica.L on three kinds ofradical scavenging ability, indicating that flavonoids from flowers of Eriobotrya.japonica.L havestrong antioxidant activity in vitro.Essential oil from flowers of Eriobotrya.japonica.L was obtained by the method ofultrasonic-assisted extraction. The extraction process was optimized by the orthogonal design onthe basis of single-factor experiment, the optimum technological conditions were: solventvolume was200mL, extraction time was60minutes, extraction times was3, extractiontemperature was35℃, under these conditions, the extraction rate of essential oil from flowers ofEriobotrya.japonica.L was (11.583±0.035)%.
