| In this paper, the activity of essential oil and liposoluble antioxidant components of rosemary have been studied after extracting, isolating and purifying essential oil and antioxidant components from rosemary, to implement the purpose of comprehensive development and utilization of rosemary resources. The main research contents were as follows:1ã€To dry the fresh leaves of Rosemary by three drying methods:insolation, drying at55℃and drying in the shade. The results showed that the essential oil yield of Rosemary leaves by insolation and drying at55℃had some loss. The essential oil yield of Rosemary leaves of drying in the shade remained intact, and the average essential oil yield was2.29%.2ã€In the research on the extraction technology of essential oil from Rosemary through steam distillation, based on the results of the single factor experiments, the orthogonal test was adopted to test the three important effects. The results showed that, the optimum extraction conditions of Rosemary essential oil were:solvent/solid ratio12:1, immersion time0h and distillation3h. The average essential oil yield was2.29%.3ã€The results showed that, among the three given kind of bacteria, the essential oil of Rosemary has a certain inhibiting effect on E.coli and Bac.subtilis, which was positively associated with the concentration of the essential oil. As to the three given kind of fungi, the same result took place when essential oil was added to the Asp.niger. All the experiment indicated that the essential oil of Rosemary has a relatively high inhibiting effect on bacteria and fungi.4ã€In the research on the content of total phenolic acids of Rosemary antioxidants determined by Folin-Phenol colorimetry, the results showed that, the best colorimetric conditions under765nm wavelength were:the volume ratio between Folin-Phenol reagent and10%Na2CO3solution was1:3, the dosage of Folin-Phenol reagent was1.5mL, the best temperature of colorimetric system was25℃, reaction time was2h.5ã€In the research on the extraction technology of Rosemary antioxidants used anhydrous ethanol as extraction solvent by reflux extraction, based on the results of the single factor experiments, the orthogonal test was adopted to test the four important effects. The results showed that, the optimum extraction conditions of Rosemary antioxidants were: ethanol concentration90%, solid/solvent ratio1:12, extraction temperature75℃extraction time2h and extraction2times. The average yield of extracts was19.10%, and the content of total phenolic acids was11.64%.6ã€Under the specified chromatographic conditions, characterized by retention time and quantified by peak area, external standard method was used to determine the content of active ingredients of Rosemary liposoluble antioxidants through HPLC. The results showed that, respectively, the content of Carnosic Acid and Carnosol was30.66%and5.12%。7ã€H1020macroporous absorbent resin was used to purify Rosemary liposoluble antioxidants. The results showed that, the adsorption isotherm curve of H1020resin was S style, the saturated adsorption capacity of H1020resin was5.337mg/g(dry resin), saturated adsorption time was3h and the suitable strippant solvent was90%ethanol solution. After dynamic desorption, the content of Rosemary liposoluble total phenolic acids increased from19.51%to56.17%and the ICso value of Rosemary liposoluble antioxidants that be purified was12.60μg/mL. |
