| Objective: To investigate the expression of HMGA1and Ezrin in laryngealcarcinoma and analyze their clinical significance and the relationship betweenHMGA1and Ezrin.Methods:The expression of proteins of HMGA1and Ezrin was analyzedsemi-quantitatively by SP Immunohistochemistry staining in50cases of laryngealcarcinoma.30cases of laryngeal polyp and30cases of atypical hyperplasia oflaryngeal were studied as controls. The expression mRNA of mRNA of HMGA1andEzrin were analyzed by real-time PCR in30cases of laryngeal carcinoma,30cases ofthe cancer cut margin were studied as controls. The expression mRNA of proteins ofHMGA1and Ezrin were analyzed by real-time PCR in30cases of laryngealcarcinoma,30cases of the cancer cut margin were studied as controls.Results:(1)The positive rates of HMGA1protein were68.0%(34/50),53.0%(16/30) and13.0%(4/30) in laryngeal carcinoma, laryngeal polyp and30cases ofatypical hyperplasia of laryngeal, and that of Ezrin protein were64.0%(32/50),50.0%(15/30) and23.3%(7/30), respectively. The expression of proteins of HMGA1andezrin were related to clinical stages(P <0.05), pathologic stages(P <0.05) and lymphnode metastasis(P <0.05), but not related to patients’age, gender or tumorplace(P>0.05).(2) HMGA1and ezrin mRNA were both expressed in laryngealcarcinoma and in cut margin of the cancer tissue(100%),The expression of HMGA1and Ezrin proteins were related to clinical stages(P <0.05), pathologic stages(P <0.05)and lymph node metastasis(P <0.05), but not related to patients’age, gender or tumorplace(P>0.05)(3) HMGA1and ezrin Proteins were both expressed inlaryngeal carcinoma and in cut margin of the cancer tissue by Western blot(100%). The expression of HMGA1and ezrin related to clinical stages(P <0.05), pathologicstages(P <0.05) and lymph node metastasis(P <0.05), but not related to patients’age,gender or tumor place(P>0.05)(4)There are positive correlations between theproteins and mRNA expression of HMGA1and Ezrin.(r=0.385, P=0.000, by realtime PCR;x2=7.172,r=0.379,P=0.012,by IHC; Western blot,r=0.412,P=0.000)Conclusion:(1)The expression of HMGA1and Ezrin protein and mRNA isabnormalities, the expression is higher than normal laryngeal tissue; In the processof malignant transformation of laryngeal atypical hyperplasia, with theincrease in severity, the expression positive rate of HMGA1and Ezrin increasedgradually;(2) HMGA1and Ezrin protein and mRNA and lymph nodemetastasis, pathological grade and clinical stage, While there is no significantcorrelation with the patient’sage,gender,tumor-location,both in the laryngeal tissues,there is a positive correlation;(3) HMGA1and Ezrin expression in laryngealcarcinoma increased the occurrence and the development process of laryngealcancer, They may play a role as the laryngeal predictthe biological markers ofMetastasis and prognosis. |
