Expression Of Integrin β3，Survivin In CinⅢ With Cervical Squmous Carcinoma And Role In Appraising The Efficacy Of Neoadjuvant Chemotherapy For Cervical Squmous Carcinoma

OBJECTIVEBy study expression of Integrin β3, Survivin in CINⅢ and cervical squmous Carcinomabefore and after neoadjuvant chemotherapy,to explore the relationship between the two indexesand the clinical and pathologic features of cervical disease, to assess the role in appraising theefficacy of neoadjuvant chemotherapy for cervical cancer.METHODS1.CIN Ⅲ of20cases and Cervical squmous Carcinoma of62cases (31cases,before and afterNeoadjuvant chemotherapy to drawn the materials),followed FIGO2000clinical standard:Therewere5cases of ⅠB,11cases of ⅡA,15cases of Ⅱ B,except that,10cases of normal cervicaltissues for control group.2.We adopted Two-steps immunohistochemical staiming method to detect the expression ofIntegrinβ3and Survivin in normal cervical tissues,CINⅢ tissue and Cervical squmouscarcinoma(before and after NACT).Positive expression of Integrinβ3can be scaned as browmgranular substance in membrance or cytoplasm;Expression of survivin can be checked as yellowto browm paticles,mainly in cytoplasm and nucleus.If the cytoplasm or nucleus has no differentwith background,it was negative effect.3.The image analysis software of Image-Pro Plus6.0was utilized for semi-quantitiveanalysis of Integrinβ3and Survivin expression,in a form of optic density (OD) withmean±standard deviation.SPSS17.0was used for statistical analysis,including t Test,Fisher,sExact Test,Variance analysis and Spearman rank correlations analysis.RESULTS1.The expression of Integinβ3and Survivin gradually enhanced from CINⅢ to cervicalsqumous Carcinoma,which have statistical significance （F=12.731,P=0.000;F=12.877,P=0.000）.There is a relation between Integrinβ3and clinical stages、organizationdifferentiation and lymph node status（P＜0.05）,however it was not relevant with the patient age(P>0.05). Survivin was related with clinical stages,organization differentiation and lymph nodemetastasis of cervical squamous cell carcinoma(P＜0.05）,has no connection with patient age (P ＞0.05).2.Both expression of Integrinβ3and Survivin were significantly reduced after neoadjuvantchemotherapy（t=2.112,P＜0.05;t=2.118，P＜0.05).3.In clinical effective group(21cases),both expression of Integrinβ3and Survivin weresignificantly reduced after chemotherapy（t=2.742, P<0.05;t=2.648, P<0.05）; No significantdecline in10clinically stable group after NACT. The decline proportion of Integrinβ3andSurvivin were obviously higher in clinical stable group than to the effective(CR+PR) group(P=0.040).4.The complete remission rate of Integrin β3and Survivin with negative was significantlyhigher than the corresponding with positive.5.The expression of Integrinβ3decreased after NACT in the high-middle differentiationgroup（P <0.05）.The expression of Integrinβ3in Cervical squamous carcinoma after NACT wasnot associated with age,clinical stage，low differentiation and lymphatic metasitasis status(P>0.05).The expression of Survivin decreased after NACT used in the high-middle differentiationgroup with no lymphatic metasitasis group （P <0.05），The positive expression change ofSurvivin in cervical squamous carcinoma before and after NACT was not associated with age ofpatients,clinical stage，low differentiation and lymphatic metasitasis (P>0.05).CONCLUSION1.The positive expression of Integrinβ3rise from CINⅢ to cervical squmousCarcinoma.There is a relation between Integrinβ3and clinical stages,organization differentiationand lymph node status.2.The positive expression of Survivin rise from CINⅢ to cervical squmous Carcinoma.Thepositive expression of Survivin has connection with clinical stages,organization differentiationand lymph node status.3.Significantly decreased expression of Integrin β3and Survivin after NACT, indicated thatthey are closely related to the efficacy of neoadjuvant chemotherapy for cervical cancer.
