In this paper, the effective ingredients in Paeonia lactiflora Pall.,Dracocephalum moldevica L.and Panax notoginseng (Burk) F.H.Chen were selectedas evaluation index to optimize the purified technological parameter of resin. Then theinfluence of temperature, bubbles and ratio of diameter to height in the practicalapplication of the macroporous resin were studied basis on the best technologicalparameter. The conclusions have very significance on the practical application ofmacroporous resin in the industrialization of traditional Chinese medicine.To study the purification technology by macroporous resin of Paeonia lactifloraPall., After investigated the adsorptive property of five kinds of macroporousadsorptive resins, The AB-8type of resins was selected by indicative constituentspaeoniflorin. The sample concentration was0.5g/mL, the absorptive flow rate was1.0mL/min, the ratio of diameter and height was1:9, the sample volume was1.5volume and the eluant was8volume of50%ethanol. We have studied the effect ofpurification of paeoniflorin by the Tandem column chromatography on the bestpurified technological parameter. Compared to the normal column chromatography,the Adsorption ratio of paeoniflorin increased6.2%; the Eluation ratio have increased3.4%. In the investigated of temperature and bubbles, compared to room temperatureAdsorption ratio of paeoniflorin have increased6.4%at40℃, and the elutionvolume have decreased2volume. The exist of bubbles make the Adsorption ratio ofmacroporous resin decreased6.8%, the Eluation ratio have decreased3.2%.In the study of the purified technological by macroporous resin ofDracocephalum moldevica L.,The UV methods for the determination of totalflavonoids was established; Through the screening of six types of macroporous resin,The AB-8type of resins was selected by tilianin and total flavonoids, The sampleconcentration was0.1g/mL, the absorptive flow rate was1.0mL/min, the ratio ofdiameter and height was1:9, the sample volume was1.25volume and the eluant was8volume of50%ethanol. We have studied the effect of tilianin and total flavonoidsby the Tandem column chromatography on the best purified technological parameter.Compared to the normal resin column, the Adsorption ratio of tilianin and totalflavonoids increased10.8%and13.3%; the Eluation ratio have increased6.5%and2.6%. In the investigated of temperature and bubbles, compared to room temperature Adsorption ratio of tilianin and total flavonoids have increased12.7%and10.0%at40℃,and the elution volume have decreased2volume. The exist of bubbles makethe Adsorption ratio of tilianin and total flavonoids by macroporous resin decreased14.5%and4.7%, the Eluation ratio have decreased3.0%and1.8%.In the study of the purified technological by macroporous resin of Panaxnotoginseng (Burk) F.H.Chen, The UV methods for the determination of totalsaponins was established; Throug h the screening of seven types of macroporous resin,The AB-8type of resins was selected by total saponins, The sample concentration was0.1g/mL, the absorptive flow rate was1.0mL/min, the ratio of diameter and heightwas1:9, the sample volume was1volume and the eluant was8volume of50%ethanol. We have studied the effect of total saponins of paeoniflorin of the “Tandemtower plate resin column” with the best purified technological parameter. Comparedto the normal resin column, the Adsorption ratio of total saponins increased9.9%; theEluation ratio have increased5.7%. In the investigated of temperature and bubbles,compared to room temperature Adsorption ratio of total saponins have increased18.1%at40℃, and the elution volume have decreased2volume. The exist ofbubbles make the Adsorption ratio of total saponins by macroporous resin decreased11.4%, the Eluation ratio have decreased2.4%.
