| Objective:By detecting the expression of caspase-3 and caspase-7 in CA3 area of the hippocampus and the cortex of the post-status epilepticus of temporal lobe epilepsy induced by electrical stimulation of the amygdala in rat. Discussion if there are the apoptosis neurons in the hippocampus of the post-status epilepticus of temporal lobe epilepsy induced by electrical stimulation of the amygdala in rat. And analyse the relationship between the apoptosis of the nerve cell and temporal lobe epilepsy.Methods:Preparetions electrode in the right amygdala of the laboratory rat. Status epilepticus which was induced by a 20 min stimulation of the amygdale. Stimulated rats were monitored whit a video recording system for a 2-months follow-up period to the appearance of SRS. Recorded rats attack access fit with Racine method. Experiment-SRS group appear is the rats with three times or more III level attacks or at least 1 time IV, V level attack. Experiment-NSRS group is the rats do not appear seizures or appear not achieve the above standard. The control group is the rats just been given preparetions electrode and not been given SE induced. Then immunohistochemical and western blotting are used to investigat the expression of caspase-3 and caspase-7 in bilateral hipp-ocampal CA3 area and the cortex of the control group, the experiment-SRS group and the experiment-NSRS rats.Results:Caspase-3 high expression was examined in the hippocampal CA3 area and low expression in the cortex of the experiment-SRS group and caspase-7 low expression in the hippocampal CA3 area of the experiment-NSRS group by using western blotting and immunohistochemistry. In contrast, caspase-3 expression was not examined in the hippocampal CA3 area and the cortex of the control group by using western blotting and immunohistochemistry.Caspase-7 low expression was examined in the hippocampal CA3 area of the experiment-SRS group and caspase-7 expression was not examined in the hippocampal CA3 area of the experiment-NSRS and e control group and the cortex of the control group, the experiment-SRS group and the experiment-NSRS rats by using western blotting and immunohistochemistry.Conclusion:This study has examined caspase-3 high expression and caspase-7 low expression in the hippocampal CA3 area of the experiment-SRS group rats induced by electrical stimulation of the amygdala. There were apoptosis neurons in the hippocampal CA3 area of the experiment-SRS group rats induced by electrical stimulation of the amygdala, and apoptosis effectors were caspase-3 and caspase-3. caspase-3 play an important role in apoptosis neurons in the hippocampal CA3 area. Ischemia and hypoxia damage induced by seizures maybe is the cause of apoptosis neurons in the hippocampal CA3 area of the SE modle. |
