Objective:The inflammatory basis of atherosclerosis has establishment. IL-8 is a powerful neutrophil chemotactic factor. In the scientific literature, there is sufficient evidence to support beyond any doubt the involvement of IL-8 in the establishment and preservation of the inflammatory micro-environment of the insulted vascular wall. Our research aims to study the relationship between the plasma level of interleukin-8 (IL-8) and the polymorphism of IL-8 single nucleotide with the susceptibility to coronary artery disease(CAD).Method:We included 244 CAD patients and 170 healthy controls. Their peripheral blood were divided into plasma and blood cells. The plasma levels of IL-8 were detected by enzyme-linked immunosorbent assay. Polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) was used to detect the genotype of IL-8-251 in CAD cases and control subjects. To evaluate chosen samples to fit random principle, the SNP of subjects were computed by haploview software:genotype and allele frequencies between groups compared by X 2 test to identify related polymorphisms of IL-8 gene with primary gouty arthritis. The OR values (odds ratio)between the two groups were Calculated and the relative risk of Different genotypes and alleles were analyzed.Result:1、IL-8 plasma levels between cases and controlsCompared with that of the control subjects, the plasma levels of IL-8 of the CAD cases were significantly increased (p<0.05).The difference is significant in spite of the classical vascular disease risk factors(OR=1.58,95%CI:1.19-2.09,P=0.002).2、Genotype and allele frequencies in cases and controls The overall distribution of IL-8-251 genotype and allelotype in CAD patients and controls were not significantly different(p>0.05). However, a statistically significant lower frequency of the AA genotypes was observed in acute coronary syndromes patients compared to other patients (OR=0.43,95%CI:0.2-0.97,P=0.04).Conclusion:Our results suggest that plasma level of IL-8 expression may co-relate with the susceptibility of CAD,-251A/T single nucleotide polymorphism is not associated with the susceptibility of CAD, but the genetic diversity may influences the clinical manifestation of CAD.
