Objective:To investigate the distribution of genetic polymorphism of SIRT6, PON1, HSP70 gene in Bama, to discuss the correlation between genetic polymorphisms of these three genes and longevity in Bama, and to explore the influence of the exposed factors on longevity in Bama.Methods:Based on the primitive record data during the fifth national census in 2000, demography data((?)100 years) of the Bama area was statistical analyzed. The geomagnetic field of the longevity district was measured by the population distribution of longevity to be the top three in Bama, the non-longevity district was measured by that to be the last two, and the external control district was decide by the custom which is similar as Bama in Nandan. It is collected 151 longevity people aged 90 to 110 from longevity district of Bama county. The control groups were composed by five groups. The controlⅠ-Ⅱcame from the longevity district, and the controlⅢ-Ⅴcame from the no-longevity district. The control I was composed of 101 people who were direct dependents of that in the longevity group. The control II was composed of 198 who had no-longevity family between 22 and 83 years. The controlⅢwas composed of 18 aged people between 90 and 105 years. The control IV was composed of 116 people aged between 48 and 89 years who had no-longevity family. The control V was composed of 176 people aged between 20 and 82 years who had no-longevity family in Nandan. After getting subjects’permission, the blood samples were collected and DNA was extracted from them, then detecting the polymrophisms of SIRT6, PON1, HSP70 gene with Taqman SNP Genotyping Assays. All data were performed withχ2 statistics, kruskal-wallis test and logistic regression analysis by using the software program SPSS. P< 0.05 was considered significant.Results:1.No significance diviation from Hardy-Weinberg Equilibrium (HWE) for the polymorphisms in SIRT6, PON1 and most HSP70 gene for the study population were observed.2. In the comparison of SIRT6, PON1, HSP70 polymorphism, there was no significant difference in genotype distribution, allele frequency and genotype classification between case and controlⅠ-Ⅳ(P<0.003), but differ from control V(P>0.003). 3. There is a significant difference at genotype distribution of Rs 350846 inSIRT6 gene (P< 0.05) between longevity region and non-longevity region, there is statistically significant correlations between G alleles and the possibility of non-longevity compare with CC genotype (Adjusted OR:0.586; 95%CI: 0.346~0.993; P:0.047). There is not a significant difference at genotype distribution of Rs 662 in PON1 gene (P> 0.05) between longevity region and non-longevity region, there is a statistically significant correlations between G alleles and the possibility of longevity compare with AA genotype (Adjusted OR:1.179; 95%CI:0.703~1.976). There is a significant difference at genotype distribution of Rs 2227956 of HSP70 gene (P< 0.05) between longevity region and non-longevity region, there is statistically significant correlations between T alleles and the possibility of longevity compare with CC genotype (Adjusted OR: 3.199; 95%CI:1.712~5.977; P:<0.001).4. By multivariate analyze,5 main influencing factors were ascerianed: gender (Adjusted OR:2.904; 95%CI:1.826~4.617), BMI(Adjusted OR:1.402; 95%CI:1.027~1.914), systolic pressure (Adjusted OR:0.972; 95%CI: 0.964~0.980), TC(Adjusted OR:1.549; 95%CI:1.219~1.968) and HDL(Adjusted OR:0.028; 95%CI:0.013~0.061).Conclusions:1. There are unique SIRT6、PON1、HSP70 allele distributions in Bama longevous population that differ from non-longevous population in other geographic areas.2. It is suggested that the population from longevity region carrying C-allele of Rs 350846 in SIRT6 gene, G-allele of Rs 662 in PON1 gene and T-allele of Rs 2227956 in HSP70 gene may possibly increase longevity.3. Longevity in Bama may be influenced by C-allele of Rs 350846 in SIRT6 gene, G-allele of Rs 662 in PON1gene, T-allele of Rs 2227956 in HSP70 gene and different geographic environment, which is probably one of the reasons that there is a uniform distribution of longevous population in Bama.4. Gender, BMI, systolic pressure, TC, HDL may be the influencing factors to the phenomenon of longevity in Bama.
