Objective To study on the changes and significance of the DNT cells and T lymphocytesubtype in children with acute lymphoblastic leukemia.Materials and methods1. subjects: 30 cases of ALL children and 24 normalchildren in control group. 2. testing indicators:①Staining: after grouping Children with ALL andhealthy control children , extract l ml from peripheral blood l ml then inject them into two testtubes, adding sodium citrate in two test tubes . In the two test tubes were addedantibody(CD45-FITC/CD4-PE/CD8-ECD/CDPC5andCD45-FITC/CD56-PE/CD19-ECD/CD3PC5) 10μl respectively , then in each test tube, add 20μl blood anticoagulant ,thoroughly mixeddark at room temperature for 30 minutes.②Hemolysis:amount 100μl hemolysin withtolerance ,add into the above two tubes, and shaking and thoroughly mixed, then placed forabout 15 minutes, using a centrifuge (1200 rev / min) and centrifuged for 10 minutes, drain thesupernatant gently.③Ablution: add buffer 2m1 in the above two test tubes, mix and centrifugedagain,,deserting the upper liquid again.④Awaiting to be detected: Using pipette to take 0.5m1buffer, adding into the above test tubes, awaiting to be detected.We choose 10000 cells fromeach semple,and detecting the expression of and CD3+CD4-CD8- T,CD3 +T cells, CD4 + Tcells, CD8 + T cell by flow cytometry.3. Data processing:,unified statistical software (SPSS13.0software) testing and accurate analysis, the final results obtained was indicated bymean±standard deviation, the two the group results were compared with t test , p <0.05 for thedifference statistically significant.ResultsDNT cells、CD3 cells and CD4 cells were decreased significantly than those innormal control .which were (4.93±3.75)%VS(8.19±3.21)%(t=3.4 P<0.01)、(49.99±11.70)%VS(64.13±11.39)%(t=4.1 P<0.01)、(28.30±7.56)%VS(34.61±6.43)%(t=3.2 P<0.01) .CD8 cellswere increased significantly than those in normal control , which was (31.19±9.89)%VS(24.33±4.24)%(t=3.1 P<0.01).Conclusion The changes of DNT cells and T subsets in children with ALL suggest animmune disorder in body, which is may be related to the occurrence of ALL.
