| ObjectiveIn recent years,the incidence of prostate cancer is showing a rising trend. Prostate specificantigen(PSA) is an important tumor marker for early screening and diagnosis of prostate cancer.PSA is the prostate specific antigen, rather than the prostate cancer specific antigen. And the testresults have the high false of positive rate, while normal PSA results can not be completely ruledout prostate cancer. Therefore, looking for a sensitive, specific, safe and convenient tumormarkers of prostate cancer, is urgent needs of urology clinicians. Sarcosine, as a non-essentialamino acids of human body, they are widely present in the prostate cancer tissue and urine. Thisstudy demonstrated sarcosine can become an early diagnosis of prostate cancer and determinethe progress and prognosis of tumor markers matter.Materials and methodsCollect blood and urine samples of patients with benign prostatic hyperplasia and prostatecancer, run by LC-MS/MS platform of specific acquisition methods, sample processing andamino acid content in the samples calculated according to the specific correction factor. Differentamino acids leucine and valine belong to the non-human body, in the same sample test contains10pmol are isoleucine and norvaline. Is isoleucine introduced in the sample pretreatment stepfor the inspection and the determination results of each amino acid amino acid recovery rate afterthe precipitation, the calibration is different leucine recovery. The final data is analysised bySPSS17.0.Result1ã€The age and prostate volume of benign prostatic hyperplasia and prostate cancer patientswere showed no significant statistical difference;2ã€The two indicators of tPSA and fPSA have significant differences. Quantitative value ofthe index sarcosine levels in blood samples of patients with benign prostatic hyperplasia and prostate cancer patients and its content was no difference (P=0.172), while the two types ofpatients with urine sarcosine levels in the quantitative values are different (P <0.05).3ã€With Gleason score/degree of the prostate cancer, urine sarcosine levels have obviousdifferences(P<0.01). The lower the degree of differentiation (the higher the degree ofmalignancy), the higher urine sarcosine levels.4ã€About sarcosine levels in urine, prostate cancer patients with bone metastases wassignificantly increased compared with non-bone metastases(P=0.007).5ã€Relevant data,16cases of prostate cancer and20cases of patients with benign prostatichyperplasia, urinary sarcosine content to take a specific cutoff, sensitivity (Se) of0.88,specificity (Sp)0.85.ConclusionThe quantitation detection of sarcosine in urine could be applied to the screen of the earlyprostate cancer; and the same time, it can be used to predict the pathological grade of prostatecancer and the progress of tumor; Finally,it was a useful tumor marker which could guide theprognostic of the prostate cancer. |
