【Objective】 To discuss the abnormity of bone marrow microenvironment of psoriasispatients by comparing the level of TGF-β1、SCF、KGF and TNF-α secreted by bone marrowstromal cells in patients and control.【Methods】 Separate bone marrow mononuclear cells of patients and control by densitygradient centrifugating. Bone marrow stromal cells were cultured with the method of adherent.When the cells were cultured for three passages and72h, bone marrow stromal cells andsupernatant liquid were collected. The phenotypes of cells were identified by flow cytometry andthe level of TGF-β1、SCF、KGF and TNF-α were detected by ELISA kits. psoriasis severityusing PASI seore method,the level of ytokines and its relationshipWith diseases everity usingPears one or relation analysis.【Results】(1)90%Cell surface antigen is high positive rate of CD29, CD45、CD34、HLA-DR is negative.The purity of bone marrow stromal cells was over90%.(2)The level ofSCF in patients was significantly higher than that in control(P<0.05),The level of TNF-α inpatients was significantly lower than that in contro(lP<0.05). the level of KGF and TGF-β1wasno significant difference between the two groups(P>0.05). And the psoriasis patient group SCF,TNF-α, KGF, TGF-β1secretion and the severity of the disease there is no significant relationship(P>0.05).【Conclusions】 Some of the cytokines,such as SCF and TNF-α, are abnormal which secretedby psoriatic bone marrow stromal cells. Which shows that psoriatic bone marrowmicroenvironment is abnormal.
