Objective: The purpose of this study is to elucidate the role of Beclinl andLC3Ⅱ in the genesis and progression of cervical cancer by detecting the Beclin1andLC3Ⅱ expression in different cervical tissue;to elucidate the relationship betweenBeclin1and LC3Ⅱ in the genesis and progression of cervical cancer.Methods: This study comprised76patients,including50cases of cervicalcancer and26cases of normal cervix tissue.1. Study group:50patients were entered with FIGO stage I and Ⅱ of cervicalsquamous cell carcinoma underwent initial treatment for radical hysterectomy andbilateral pelvic1ymphadenectomy in the Second Affiliated Hospital of SoochowUniversity from Jan.2007to Dec.2008.The age of patients ranged from20-68yearsold, with a median of42years old.36cases were I stage (72%),14cases were Ⅱstage (14%).2. Control group:26cases were all subjected to complete hysterectomy due tohysteromyoma during the same period of time.The normal cervical tissue of thesecases were detected. The age of patients ranged from36-55years old, with a medianof43years old.All above tissues were all fixed in formalin and embedded with paraffin.Beclin1and LC3Ⅱ were stained by immunohistochemistry. Immunohistochemicalstaining results were counted semiquantitively.Statistical analysis was performedusing SPSS17.0. Results:1. Expression of Beclinl and LC3Ⅱ in cervical tissueThe positive expression rate of Beclin1in normal cervix and cervical squamouscell carcinoma was96.2%and28%respectively. Significant difference was observedamong them (P=0.000).The positive expression rate of LC3Ⅱ in normal cervix and cervical squamouscell carcinoma was76.9%and26%respectively. Significant difference was observedamong them (P=0.000).2. The relationship between Beclin1and LC3Ⅱ with clinicopathlogical factors.The expression of Beclin1and LC3Ⅱ were not associated with age, tumor size,tumour differentiation, FIGO stage, depth of invasion, vascular invasion, parametrialinvolvement and pelvic lymphnode metastasis (P>0.05).Univariate analysis of clinicopathlogical factors revealed that vascular invasion(P=0.0009), pelvic lymphnode metastasis (P=0.002) and Beclin1(P=0.0341)wererisk factors of overall survival (OS). Multivariate analysis showed that vascularinvasion (P=0.036), pelvic lymphnode metastasis (P=0.016)were the most powerfulindependent prognostic factors for overall survival (OS).3. The relationship between Beclinl and LC3ⅡThe expression of Beclin1were not correlated with LC3Ⅱ expression(P=0.329).Conclusions:1. Beclin1and LC3Ⅱ level significantly decrease in cervical squamous cellcarcinoma than that in normal cervical tissue, which implys that they may play a rolein carcinogenesis and development of cervical squamous cell carcinoma.2. The expression of Beclin1and LC3Ⅱ were not associated with age, tumorsize, tumour differentiation, FIGO stage, depth of invasion, vascular invasion,parametrial involvement and pelvic lymphnode metastasis. 3. The vascular invasion, pelvic lymphnode metastasis and Beclin1were riskfactors of overall survival (OS) for cervical squamous cell carcinoma.4. No significant correlation is observed between Beclinl and LC3Ⅱ in thestudy.5. The vascular invasion, pelvic lymphnode metastasis were the most powerfulindependent prognostic factors for overall survival (OS).
