Research Background:Cervical cancer is the most common gynecologic malignant tumor, currentlyconsidered the main factors for cervical cancer is the human papilloma virus (Humanpapillomoviruses,HPV) infection, Especially persistent high-risk HPV infection. Withthe continuous improvement of the level of medical treatment, cervical cancer isbasically able to do early detection and early treatment. But with the changes in thesocial life, the HPV infection rates continue to rise, the incidence of cervical cancerare getting younger and younger. Cervical cancer has a serious impact on the health ofwomen,which is one cause of cancer deaths in women. Autophagy is a process thatcellular via lysosome degradation of damaged or aging intracellular organelles and theof macromolecular substances, at the same time provides energy for cell recycling. Inrecent years, the relationship between the tumor and autophagy is gradually deepening,through the regulation of autophagy activity affect tumor cell proliferation and thusplay a therapeutic effect on tumor. Expression of ARHI and Beclin1were decreasedin a variety of malignant cells, both of autophagy-related genes, by regulating theactivity of autophagy play a important role in tumorigenesis and development. In thisstudy, we using immunohistochemical methods to detected autophagy gene ARHI andBeclin1expression in cervical cancer, and analysis of the relationship betweenARHI、beclin1and some clinical parameters (histological grade, clinical stage, lymphnode metastasis) of cervical cancer. Explore the impact of autophagy genes ARHI andBeclin1on cervical cancer and look forward to be able to provide a new starting pointfor the treatment of cervical cancer.Materials and Methods:All of patients were randomly selected for their initial treatment derived fromGynecology department of the second hospital of Jilin University from September 2011to October2012, including12cases with normal cervix,15cases withCINⅡ-Ⅲ,50cases with cervical cancer (including34cases without lymph nodemetastasis and16cases with lymph node metastasis). The pathological diagnosis ofall patients with cervical cancer are squamous cell carcinoma,and confirmed by thepostoperative paraffin sections. All77cases of specimens were stained byimmunohistochemical method SP,and the main steps in reference to the kitinstructions. The statistical software SPSS for Windows (version17.0; SPSS) wasused to perform the statistical analysis. x2inspection was used in counting data,pearson analysis was used in correlation. Statistical results of P<0.05was consideredstatistically significant.Results:1、The expression of ARHI in normal cervical tissue、 CIN and cervical cancerwere91.67%(11/12)、73.33%(11/15)、52.0%(26/50). Comparison between the threegroups, the difference was statistically significant (P <0.05). The difference betweennormal cervical tissue and CIN was statistically significant (P>0.05), The differencebetween cervical tissue and cervical cancer was statistically significant (P<0.05),Thedifference between cervical cancer and CIN was statistically significant (P>0.05).2、The positive rate of32cases of I stage were65.63%(21/32),the positive rateof18cases of II stage were27.78%(5/18), the difference was significant instage(P<0.05);The difference between high differentiation (17/26,65.38) andmiddle-low differentiation(9/24,37.5%) was significant difference (P<0.05);Thepositive rate of lymph node metastasis were43.75%(7/16),the positiverate of withoutlymph node metastasis were55.88%(19/34),the difference was no significant(P>0.05).3、The expression of Beclin1in normal cervical tissue、CIN and cervical cancerwere100%(12/12)、60.0%(9/15)、26.0%(13/50). Comparison between the threegroups, the difference was statistically significant (P <0.05). The difference betweennormal cervical tissue and CIN、cervical tissue and cervical cancer and cervicalcancer and CIN were all statistically significant (P<0.05). 4、The positive rate of I stage were28.13%(9/32),the positive rate of II stage were22.22%(5/18),the difference was no significant in stage(P>0.05); The positive rate ofhigh differentiation were23.07%,(6/26),and the positive rate of middle-lowdifferentiation were29.17%,(7/24),the difference was no significant(P>0.05);Thepositive rate of lymph node metastasis were12.5%(2/16),the positiverate of withoutlymph node metastasis were32.35%(11/34),the difference was no significant(P>0.05).5、The expression of ARHI and Beclin1in cervical cancer has a positivecorrelation, r=0.387,p=0.006.Conclusion:1、The expression of ARHI and Beclin1were gradually reduce in normal cervicaltissue、CIN and cervical cancer, prompted the two may be involved in the occurrenceof cervical cancer.2、The expression of ARHI was related to clinical stage of cervical cancer,prompted the ARHI may be involved in the development of cervical cancer;Theexpression of ARHI was related to Tissue differentiation degree,prompted the ARHImay be involved in tissue differentiation and Level of malignant in cervical cancer.3、The expression of ARHI and Beclin1in cervical cancer has a positivecorrelation(r=0.387, p=0.006),They may be through a common mechanism orinteractions play an impact in the development of cervical cancer.
