| Diabetes is one of the most common chronic diseases characterized by hyperglycemia with endocrine and metabolic disorders caused by insulin secretion and utilization disorder. Incidence of diabetes has increased year by year, and it has become one of the most prominent problems in the worldwide, a-glucosidase inhibitors are novel per os drugs being used to control blood sugar. At present, acarbose, voglibose and miglitol are used for clinical treatment generally. At present, acarbose. voglibose and miglitol are generally used in clinic. They can control the blood sugar of diabetes patients, prevent the condition from deteriorating, and reduce diabetes complications. Mulberry leave is a traditional chinese medicine herb. Some research results have showed that its leaf contains a large amount of a-glucosidase inhibitors. But there is scarcely any systematic study on their convenient and efficient extract method.We used the a-glucosidase inhibitor screening model in vitro to detect the activity of a-glucosidase which is inhibited by mulberry leaf extracts with different methods, to ensure the method to obtain the extracts with high inhibition. The result showed that the optimum extraction method was that mulberry leaf was extracted with boiling water for30min, and the supernatant of the extract solution was further extracted with water-ethyl acetate, then aqueous phase was treated through D101macroporous resin, elutriated it with water, the eluate was treated through polyamide column, elutriated it with water, the eluent was treated through G25gel column, and elutriated it with water again, and collected the eluates as50ml, we get S004S-2and S004S-3, S004S-5, S004S-7, S004S-11. We detecting IC50of the four extracts.The results showed that they could inhibit a-glucosidase significantly.We examined the effects of the four extracts on blood glucose and other biochemical parameters of diabetic mice induced with alloxan. We kept the mice with the feed containing a certain proportion of acarbose or extract. We found that the extracts could significantly decreased blood glucose of diabetic mice. The blood glucose closeed to normal levels after30days. Their weights increased significantly. The extracts could significantly meliorate blood lipids, kidney-body weight ratio and lipid peroxidation. |
