| Objective To investigate the effect of the glucagon-like Peptide1on the gastric emptying in dorsal vagal complex (DVC) of streptozotocin-induced diabetic rats,and to study the related mechinisms of thses effects.Methods Diabetes was induced by intrapritoneal injection of streptozotocin (60mg/kg) in male Wistar rats.All rats were surgically implanted with indwelling cannulas in the dorsal vagal complex,and saline or GLP-1(1μg) in0.5μl saline was delivered to the DVC.Gastric emptying of phenol red solution and the plasma GLP-1levels measured by Enzyme-Linked Immunosorbent Assay (ELISA),were assessed two weeks of the STZ injection,while the GLP-1receptor(GLP-1R) mRNA of the gastric antrum and fundus were examined by means of reverse transcriptase-mediated polymerase chain reaction (RT-PCR).Results Two weeks after streptozotocin,gastric emptying was enhanced (72±11%) compared with control rats (35±8%, p<0.05), but the plasma GLP-1level did not change significantly. GLP-1treatment in the DVC attenuated accelerated gastric emptying significantly (39±12%)and increases the plasma GLP-1(15.35±2.21pmol/L) of the diabetic rats (10.78±2.06pmol/L,p<0.05),while the GLP-1R mRNA of the gastric antrum/β-actin (1.33±0.22) was also increased compared with the diabetic rats (1.10±0.14,p<0.05).There was not any significant change of the GLP-1R mRNA of the gastric fundus in our experiment.Conclusion GLP-1treatment in the DVC can inhibit the accumulated gastric emptying in the early stages of diabetes rats.this maybe achieved through increasing the level of the plasma GLP-1and the GLP-1R mRNA of the gastric antrum. |
