ObjectiveTo explore the types and frequencies of chromosome aberrations in spontaneous abortion material and assess the values of FISH in the detection.Methods107 samples were collected in this study. Conventional karyotype analysis was carried out after cell culturing. The interphase nuclei of 107 abortion products were detected by FISH using original DNA probes for chromosomes 13,16,18,21,22, X, Y. The detection results of FISH were compared with chromosomal karyotype analysis.Results(1) 76 samples in 107 cases were successfully cultured. The detection rates of G-banding (71.03%) were significantly lower than the FISH (100%). The statistical difference between the two methods was found (X=36.25, P< 0.05).(2) 35 abnormal cases were detected by the conventional cytogenetic methods while 48 cases were diagnosed by FISH. Overall,46.73% of miscarriages were found to be chromosome aberrations among all cases.(3) The predominant chromosomal aberrations are trisomies (40.00%), triploidy (28.00%) and monosomy X (20.00%).ConclusionChromosomal aberrations could be detected precisely and rapidly by FISH. While FISH allows analysis of large numbers of interphase nuclei, it is an efficient tool to identify mosaicism. Chromosomal abnormality is the main reason of spontaneous abortion.
